Summary: Neospora caninum and Toxoplasma gondii are major concerns in the livestock industry, leading to economic losses and reproductive failure in ruminants. Current knowledge gaps exist in understanding the host-parasite interactions in the placenta, hindering the development of effective vaccines and treatments. Utilizing ruminant animal models, in vitro systems, and placental explants can provide insights into pathogenic mechanisms and host immune responses, but each has limitations that need to be addressed for further research and development.

Summary: This study documented the antibody responses, tissue cyst burden, and mouse virulence of Toxoplasma gondii strains isolated from different hosts in China. All T. gondii strains were found to form tissue cysts in the brains of mice, and the presence of tissue cysts positively correlated with antibody titers against T. gondii. These results may assist in the diagnosis and characterization of T. gondii isolates.

Summary: This study used the Oxford Nanopore Minion platform to generate near-complete de novo genome assemblies for multiple strains of Toxoplasma gondii and Neospora caninum, significantly improving genome contiguity and resolving hundreds of tandem repeats. Although both species retained a 13-chromosome karyotype, extensive chromosome-scale rearrangements were found to have occurred in both Toxoplasma gondii and Neospora caninum since their most recent common ancestry.

Establishment and validation of a guinea pig model for human congenital toxoplasmosis

Summary: This study investigated the impact of T. gondii infection dose, duration of infection, and gestational stage at infection on guinea pigs to establish them as a suitable model for human congenital toxoplasmosis. The study found that infection dose, gestational stage, and duration of infection play crucial roles in the survival rate of dams, fate of offspring, and integrity of offspring brain. These findings contribute to using guinea pigs as an animal model for studying pathogenesis, diagnosis, therapy, and prognosis of congenital toxoplasmosis.

Summary: The study identified significant intra- and inter-genotype virulence differences in Spanish T. gondii isolates, which were not reflected by the genetic characterization using currently described molecular markers. Isolates TgShSp16 (#3) and TgShSp24 (#2) presented higher degrees of virulence, and there were significant differences in invasion rates and tachyzoite yield at 72 h post-inoculation between isolates.
