Heme protects Pseudomonas aeruginosa and Staphylococcus aureus from calprotectin-induced iron starvation

Emily M. Zygiel et al.

Summary: This study reveals that Pseudomonas aeruginosa upregulates heme uptake machinery in response to the presence of the innate immune protein calprotectin (CP), which inhibits iron uptake. Furthermore, heme is found to protect P. aeruginosa from CP-mediated inhibition of iron uptake and iron-starvation responses. Similar results are observed with Staphylococcus aureus, suggesting the importance of heme utilization for bacterial adaptation to host iron-withholding strategies.

Heme-Dependent Siderophore Utilization Promotes Iron-Restricted Growth of the Staphylococcus aureus hemB Small-Colony Variant

Izabela Z. Batko et al.

Summary: Respiration-deficient Staphylococcus aureus small-colony variants (SCVs) require iron for persistent infections, yet it is unknown how SCVs obtain iron. HemB mutants of S. aureus utilize hemin for iron acquisition but are defective in siderophore-mediated iron acquisition, highlighting the importance of hemin as both an iron source and nutrient to promote siderophore utilization in SCVs.

A High-Throughput Method for Identifying Novel Genes That Influence Metabolic Pathways Reveals New Iron and Heme Regulation in Pseudomonas aeruginosa

David G. Glanville et al.

Summary: Heme is essential for the survival of most life on earth, including bacterial pathogens like Pseudomonas aeruginosa. By using a novel approach called Met-Seq, researchers identified 188 genes that potentially influence intracellular heme levels in P. aeruginosa, shedding light on previously unknown metabolic pathways and potential therapeutic targets.

Iron Acquisition Systems of Gram-negative Bacterial Pathogens Define TonB-Dependent Pathways to Novel Antibiotics

Phillip E. Klebba et al.

Summary: Iron is essential for microbial pathogenesis and there is a competition for this metal between bacterial pathogens and hosts. Bacteria secrete siderophores to extract iron, while hosts combat bacterial iron scavenging by sequestering iron in binding proteins and ferritin. This diversity in iron uptake systems of Gram-negative bacterial pathogens highlights various mechanisms that facilitate microbial pathogenesis.
