Structural modification aimed for improving solubility of lead compounds in early phase drug discovery

Summary: Solubility is a crucial factor in early phase drug discovery, as many potent lead compounds fail to reach clinical trials due to low bioavailability caused by insufficient solubility. Improving solubility while maintaining potency is a challenging task for medicinal chemists. Poor solubility not only hinders in vitro and in vivo assays but also prolongs drug development time and increases costs. Various techniques, including physical, chemical, and miscellaneous methods, are used to enhance solubility. The medicinal chemistry approach of structural modification is versatile and can improve other pharmacokinetic/physicochemical parameters simultaneously.

Identifying novel putative ERK1/2 inhibitors via hybrid scaffold hopping -FBDD approach

Summary: In this study, novel lead molecules with putative ERK2 inhibitory potential were discovered using molecular docking and molecular dynamic simulations. Three compounds (ligand 6, 8 and 10) were identified to possess optimum pharmacodynamic and pharmacokinetic profile to be potential ERK2 inhibitors, which can be further validated via biological evaluation.

These compounds have potential for fragment-based drug design as leishmanicides, but further studies are needed to evaluate their therapeutic potential.

Summary: This study evaluated the efficacy of bioinformatics tools in predicting the structure and epitopes of the rLigBCon1-5 antigen. Predicted structure and potential B-cell and T-cell epitopes were compared with wet lab experiments, showing reliability and potential immunogenicity of the antigen. Computational approaches were found to be reliable for the serodiagnosis of leptospirosis, with IgG Dot ELISA Dipstick test and LAT serving as suitable assays for detection of porcine leptospirosis in peripheral level animal health care systems.
