Multisite phosphorylation by Cdk1 initiates delayed negative feedback to control mitotic transcription

Jonathan B. Asfaha et al.

Summary: Cell-cycle progression is driven by the phosphorylation of cyclin-dependent kinase (Cdk) substrates. The order of substrate phosphorylation depends on the rise in Cdk activity during the cell cycle and variations in substrate docking. Ndd1, a transcriptional co-activator, is phosphorylated by Cdk1 to promote mitotic cyclin gene transcription, but high Cdk1 activity leads to inhibition of gene expression and degradation of Ndd1 through multisite phosphorylation.

Cyclin B/CDK1 and Cyclin A/CDK2 phosphorylate DENR to promote mitotic protein translation and faithful cell division

Katharina Clemm von Hohenberg et al.

Summary: The cell cycle regulates translation during mitosis by phosphorylating the DENR protein, which promotes proper mitotic progression. DENR also influences the translation of specific mitotically relevant mRNAs, and its aberrant expression leads to abnormal mitosis and cell death.

Lipophagy-derived fatty acids undergo extracellular efflux via lysosomal exocytosis

Wenqi Cui et al.

Summary: The autophagic degradation of lipid droplets, termed lipophagy, is a major mechanism that contributes to lipid turnover in numerous cell types. This study demonstrates that the fusion of lysosomes to the plasma membrane is the primary route for the disposal of fatty acids derived from lipophagy. Additionally, the efflux of fatty acids and their reuptake or subsequent extracellular trafficking may play a crucial role in cell-to-cell lipid exchange and signaling.

TFEB regulates lysosomal exocytosis of tau and its loss of function exacerbates tau pathology and spreading

Yin Xu et al.

Summary: The transcription factor EB (TFEB) plays a crucial role in the lysosomal exocytosis of selected tau species, with TFEB loss causing a decrease in tau levels and accelerated spreading within neurons. This suggests that TFEB-mediated tau exocytosis acts as a clearance mechanism to reduce intracellular tau levels under pathological conditions.

Variations of intracellular density during the cell cycle arise from tip-growth regulation in fission yeast

Pascal D. Odermatt et al.

Summary: Research has found that dry-mass density in fission yeast exhibits homeostatic behavior but varies during the cell cycle, largely influenced by a constant rate of biomass synthesis, slowdown of volume growth during cell division, and rapid expansion post-cytokinesis. Density changes are exacerbated by arrests at specific cell-cycle stages, and spatially heterogeneous patterns of density suggest links with density regulation, tip growth, and intracellular osmotic pressure. The systematic density variations during the cell cycle are predominantly due to modulation of volume expansion, revealing functional consequences of density gradients and cell-cycle arrests.
