Genome-wide association study for four measures of epigenetic age acceleration and two epigenetic surrogate markers using DNA methylation data from Taiwan Biobank

Wan-Yu Lin

Summary: The study identified genetic loci associated with epigenetic aging and its surrogate markers, with a specific focus on individuals of Asian ancestry. By conducting a GWAS and prioritized subset analysis on a Taiwan Biobank cohort, the researchers replicated previously reported aging-associated loci and identified novel loci. One of the newly discovered loci, rs117530284 in the IBA57 gene, was associated with epigenetic age acceleration. This study contributes to our understanding of the genetic architecture underlying epigenetic aging in individuals of Asian ancestry.

Epigenetic aging of the demographically non-aging naked mole-rat

Csaba Kerepesi et al.

Summary: The study reveals that naked mole-rats, despite having no increased mortality with age, experience epigenetic aging. By conducting bisulfite sequencing on the blood of over 100 naked mole-rats, researchers observe age-related methylome remodeling and loss of methylome information, indicating the aging process. An epigenetic clock is developed to accurately predict the age of naked mole-rats. These animals age at a much slower rate than mice but faster than humans, consistent with their known maximum lifespans.

A mammalian methylation array for profiling methylation levels at conserved sequences

Adriana Arneson et al.

Summary: The authors developed a Mammalian Methylation Array that includes 36k well-conserved CpGs in mammals, facilitating cross-species comparisons. They annotated the conserved CpGs in > 200 species. The array allows one to measure methylation in all mammalian species, including unsequenced ones.

Multiomic profiling of the liver across diets and age in a diverse mouse population

Evan G. Williams et al.

Summary: We analyzed the liver transcriptome, proteome, and metabolome of BXD mice to study the effects of diet and aging on molecular variations and metabolic traits. We identified genes associated with longevity and found common diet-related candidate genes across datasets. Our study provides insights into causal connections between diet, aging, and metabolic traits.

Many chronological aging clocks can be found throughout the epigenome: Implications for quantifying biological aging

Hunter L. Porter et al.

Summary: The study identified predictive sites that can be used to create various epigenetic clocks, which accurately reflect biological age. However, there was only a weak association between accelerated epigenetic aging and disease, suggesting limitations in understanding the molecular biology of aging with current epigenetic clocks.

A platform for experimental precision medicine: The extended BXD mouse family

David G. Ashbrook et al.

Summary: The expansion of the BXD family of mice to 140 fully isogenic strains has created a uniquely powerful model for precision medicine, with the ability to map heritable traits with high power and precision. The BXD family, which segregates for 6 million common DNA variants, is an effective platform for testing causal modeling and predictive validation in experimental precision medicine.

Genetic associations for two biological age measures point to distinct aging phenotypes

Summary: Biological age measures outperform chronological age in predicting aging outcomes, with genetic predisposition being most strongly associated with the APOE gene. Different biological age measures show unique genetic associations and pathway enrichments, indicating they capture distinct aging domains.

The association of epigenetic clocks in brain tissue with brain pathologies and common aging phenotypes

Francine Grodstein et al.

Summary: Epigenetic clocks, calculated by combining DNA methylation states across select CpG sites to estimate biologic age, were used in brain tissue of older participants to explore aging phenotypes. The Cortical clock showed stronger associations with Alzheimer's disease pathology compared to other established clocks, and was also significantly related to clinical phenotypes such as dementia and cognitive decline.

Gene-by-environment modulation of lifespan and weight gain in the murine BXD family

Suheeta Roy et al.

Summary: Through experiments on female mice, this study found that a high-fat diet can significantly shorten lifespan. Initial body weight and weight gain are important factors affecting longevity, while the response to a high-fat diet is modulated by gene-environment interactions. Classic serum metabolites may not be strong predictors of longevity.

Genome-wide association studies identify 137 genetic loci for DNA methylation biomarkers of aging

Daniel L. McCartney et al.

Summary: This study identified 137 genome-wide significant loci, of which 113 are novel, related to epigenetic aging using DNA methylation and SNP data from over 40,000 individuals. The findings illuminate the genetic architecture underlying epigenetic aging, including shared genetic contributions with lifestyle factors and longevity.
