Functional genomic analysis delineates regulatory mechanisms of GWAS-identified bipolar disorder risk variants

Advances toward precision medicine for bipolar disorder: mechanisms & molecules

Summary: Bipolar disorder presents a significant unmet medical need, with challenges in effective treatment and prevention. Despite more than 50 years since the introduction of lithium, understanding of its mechanisms and the pathophysiology of bipolar disorder remains limited, hindering the development of new treatments. Progress has been made in understanding the genetic complexity of bipolar disorder, but effective treatments for acute bipolar depression are still lacking. New therapeutic strategies based on defined molecular mechanisms and neural circuits may offer promise for improving the treatment of bipolar disorder.

A Human-Specific Schizophrenia Risk Tandem Repeat Affects Alternative Splicing of a Human-Unique Isoform AS3MTd2d3 and Mushroom Dendritic Spine Density

Summary: Recent advances in functional genomics have led to the identification of multiple genes and isoforms associated with the genetic risk of schizophrenia, with a specific focus on the SNP rs7085104 and VNTR at 10q24.32. This study demonstrates the direct regulation of AS3MT(d2d3) mRNA expression by the VNTR, along with its association with schizophrenia risk in East Asians. Overexpression of AS3MT(d2d3) in cultured hippocampal neurons results in reduced densities of dendritic spines, highlighting its potential functional impact on neuroplasticity.
