Hepatic macrophages in liver homeostasis and diseases-diversity, plasticity and therapeutic opportunities

Yankai Wen et al.

Summary: Macrophages, key cellular components of the liver, play essential roles in maintaining liver homeostasis, injury and repair processes in liver diseases. Kupffer cells and monocyte-derived macrophages contribute significantly to tissue inflammation and injury, but exhibit different functional responses in distinct liver diseases.

Tumor-associated myeloid cells: diversity and therapeutic targeting

Alberto Mantovani et al.

Summary: Myeloid cells in tumor tissues, including tumor-associated macrophages and tumor-associated neutrophils, play crucial roles in tumor growth and invasion. In recent years, successful therapeutic approaches have been developed and tested in preclinical cancer models, with some strategies reaching clinical trials.

Liver metastasis restrains immunotherapy efficacy via macrophage-mediated T cell elimination

Jiali Yu et al.

Summary: Liver metastases diminish the efficacy of immunotherapy by siphoning activated CD8(+) T cells from systemic circulation, creating systemic immune desert. Combining liver-directed radiotherapy with immunotherapy can promote systemic antitumor immunity.

A pan-cancer single-cell transcriptional atlas of tumor infiltrating myeloid cells

Sijin Cheng et al.

Summary: This study conducted a pan-cancer analysis of single myeloid cells from 210 patients across 15 human cancer types, revealing distinct features of TIMs in different cancer types. For example, mast cells in nasopharyngeal cancer were associated with better prognosis, and pro-angiogenic TAMs showed diverse markers across different cancer types. The composition of TIMs appeared to be linked with certain features of somatic mutations and gene expressions.

Tumor-derived adenosine promotes macrophage proliferation in human hepatocellular carcinoma

Junfeng Wang et al.

Summary: Tumor-infiltrating macrophages in human hepatocellular carcinoma exhibit higher proliferative capacity, less differentiation, and are induced by tumor cell-derived adenosine and autocrine granulocyte-macrophage colony-stimulating factor (GM-CSF). Local macrophage proliferation is an important mechanism for macrophage accumulation in HCC tissues and selective modulation at the source may offer a novel strategy for cancer therapy.
