Epigenetics/Epigenomics and Prevention of Early Stages of Cancer by Isothiocyanates

Rasika Hudlikar et al.

Summary: Cancer development can take 10-50 years and may be influenced by environmental factors impinging upon genetics/epigenetics. Using relatively nontoxic chemopreventive epigenetic modifiers could be a logical approach for cancer prevention.

m6A-RNA Demethylase FTO Inhibitors Impair Self-Renewal in Glioblastoma Stem Cells

Sarah Huff et al.

Summary: N-6-methyladenosine (m(6)A) is the most abundant mRNA modification that regulates gene expression and plays a key role in various diseases. The FTO inhibitor FTO-04 has been shown to effectively inhibit neurosphere formation in glioblastoma stem cells, making it a potential new lead for treating glioblastoma.

Safety and Efficacy of Bromodomain and Extra-Terminal Inhibitors for the Treatment of Hematological Malignancies and Solid Tumors: A Systematic Study of Clinical Trials

Yanli Sun et al.

Summary: The upregulated expression of BET proteins is closely related to hematological malignancies and solid tumors. Ten BET inhibitors currently in clinical trials showed exposure-dependent thrombocytopenia, and further efforts are needed to explore optimal dosing schemes for maximizing their efficacy.META_DESCRIPTION

Achieving clinical success with BET inhibitors as anti-cancer agents

Tatiana Shorstova et al.

Summary: Transcriptional upregulation of oncogenes drives tumor progression, and targeting BET proteins with inhibitors has shown promise in suppressing oncogenic networks in tumors. Understanding the biology of BET proteins, designing better BET inhibitors, and incorporating BET inhibitors into combination therapies can enhance their efficacy in clinical settings.

Small-molecule inhibition of METTL3 as a strategy against myeloid leukaemia

Eliza Yankova et al.

Summary: The study identified a highly potent and selective catalytic inhibitor of METTL3, STM2457, which shows potential therapeutic value in treating AML. Treatment with this inhibitor leads to reduced AML growth, increased differentiation and apoptosis, and decreased m(6)A levels on leukemogenic mRNAs. In vivo pharmacological inhibition of METTL3 results in impaired engraftment and prolonged survival in mouse models of AML.

A Review of Small-Molecule Inhibitors of One-Carbon Enzymes: SHMT2 and MTHFD2 in the Spotlight

Christine R. Cuthbertson et al.

Summary: Metabolic reprogramming is a hallmark of cancer, with enzymes like SHMT2 and MTHFD2 in the folate cycle arm of the one-carbon metabolism pathway playing crucial clinical roles. This review discusses the function, regulation, and clinical relevance of SHMT2 and MTHFD2, as well as evaluating their prognostic significance across various cancers. Additionally, it provides an overview of inhibitors targeting the folate pathway, the methionine cycle, and other related enzymes for potential cancer therapy.
