Chemodynamic Therapy via Fenton and Fenton-Like Nanomaterials: Strategies and Recent Advances

Chenyang Jia et al.

Summary: Chemodynamic therapy (CDT) is a novel cancer treatment strategy that generates hydroxyl radicals in the tumor region using Fenton or Fenton-like reactions. It has advantages such as low side effects, independence from external stimulation, modulation of the tumor microenvironment, and low treatment cost. CDT can also be combined with other therapies to enhance anticancer effects.

Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Photoacoustic Imaging and Photothermal Therapy of Semiconducting Polymer Nanoparticles: Signal Amplification and Second Near-Infrared Construction

Xu Zhen et al.

Summary: The development of semiconducting polymer nanoparticles (SPNs) for photoacoustic imaging and photothermal therapy has shown promising results due to their clear structure-property relation and molecular structural tunability. Various strategies such as structure-property screening, fluorescence quenching, accelerated heat dissipation, and size-dependent heat dissipation have been discussed to amplify the PA brightness of SPNs for in vivo PA. Additionally, molecular approaches have been introduced to shift the absorption of SPNs for NIR-II PA imaging and PTT in order to improve tissue penetration depth for diagnosis and therapy.

Recent Advances in Nanomaterial-Based Nanoplatforms for Chemodynamic Cancer Therapy

Shu-Lan Li et al.

Summary: Chemodynamic therapy (CDT) uses endogenous chemical energy in the tumor microenvironment for treatment, converting hydrogen peroxide into reactive oxygen species for oncotherapy. However, limitations in the tumor microenvironment hinder CDT efficacy, and the development of nanotechnology offers potential solutions to improve CDT efficiency.

Reprogramming the Tumor Microenvironment through Second-Near-Infrared-Window Photothermal Genome Editing of PD-L1 Mediated by Supramolecular Gold Nanorods for Enhanced Cancer Immunotherapy

Honglin Tang et al.

Summary: The photothermal genome-editing strategy using a supramolecular cationic gold nanorod for CRISPR/Cas9 delivery to disrupt PD-L1 and induce immunogenic cell death (ICD) shows promising results in improving immune checkpoint blockade (ICB) therapy. This approach enhances dendritic cell conversion to T cells, promotes infiltration of cytotoxic T lymphocytes into tumors, and reprograms the immunosuppressive tumor microenvironment into an immunoactive one to inhibit primary and metastatic tumor activity with long-term immune memory effects.

An Acidity-Unlocked Magnetic Nanoplatform Enables Self-Boosting ROS Generation through Upregulation of Lactate for Imaging-Guided Highly Specific Chemodynamic Therapy

Linan Shi et al.

Summary: A novel nanoplatform was developed in this study, which can achieve high-efficient tumor-specific catalytic anticancer treatment in acidic tumor microenvironments through a cyclic amplification strategy, leading to enhanced Fenton-like reaction. Ultimately, this smart nanoplatform enables self-boosting generation of reactive oxygen species (ROS), inducing strong intracellular oxidative stress, and resulting in substantial anticancer outcomes in vivo.

Enhanced response to PD-L1 silencing by modulation of TME via balancing glucose metabolism and robust co-delivery of siRNA/Resveratrol with dual-responsive polyplexes

Li Jia et al.

Summary: Balancing glucose metabolic pathways of glycolysis and mitochondrial OXPHOS can modulate the immunosuppressive tumor microenvironment and promote immune cell infiltration and anti-tumor effects. Downregulating glycolysis while upregulating mitochondrial OXPHOS in tumor cells leads to reduced lactate production, enhanced immune cell activity, and superior anti-tumor effects in combination with PD-L1 silencing.
