Jing Xu et al.

Summary: The study investigated the role and mechanism of melatonin-loaded polymer micelles in the treatment of dry eye disease (DED). The results showed that the melatonin-loaded polymer micelles improved the solubility and biological activities of melatonin in aqueous solutions. The study also demonstrated that the micelles alleviated hyperosmolarity-induced ocular surface damage through promoting mitophagy mediated by PINK1 and may act on the MT1 receptor to treat DED.

HMGB1 downregulation in retinal pigment epithelial cells protects against diabetic retinopathy through the autophagy-lysosome pathway

Lujia Feng et al.

Summary: This study investigated the relationship between HMGB1, the macroautophagy/autophagy-lysosome pathway, and DR, finding that high glucose inhibited autophagy and diminished its degradative capacity due to lysosome membrane permeabilization. HMGB1 was involved in lysosome membrane permeabilization via the CTSB-dependent pathway in the early stages of DR.

Torin 1 alleviates impairment of TFEB-mediated lysosomal biogenesis and autophagy in TGFBI (p.G623_H626del)-linked Thiel-Behnke corneal dystrophy

Liyuan Wang et al.

Summary: Thiel-Behnke corneal dystrophy (TBCD) is caused by mutations in the TGFBI gene, and a novel mutation was identified in a TBCD pedigree. The study utilized mouse models to verify the characteristics observed in the human sample, and suggested that lysosomal dysfunction may lead to autophagic flux obstruction in TBCD.

Exosomes Derived from Mouse Adipose-Derived Mesenchymal Stem Cells Alleviate Benzalkonium Chloride-Induced Mouse Dry Eye Model via Inhibiting NLRP3 Inflammasome

Guifang Wang et al.

Summary: The study found that mouse adipose-derived mesenchymal stem cell-derived exosomes (mADSC-Exos) have a great efficacy in the treatment of dry eye syndrome. mADSC-Exos can repair corneal surface damage, inhibit cell apoptosis, reduce levels of inflammatory cytokines, and reverse inflammasome activation. Therefore, mADSC-Exos hold promise as an effective treatment for dry eye syndrome.

The cGAS-STING signaling pathway contributes to the inflammatory response and autophagy in Aspergillus fumigatus keratitis

Fang Han et al.

Summary: The cGAS-STING signaling pathway plays a crucial role in Aspergillus fumigatus keratitis, contributing to inflammatory responses. Activation of this pathway enhances pro-inflammatory cytokine production while inhibition of cGAS can alleviate the severity of the disease. Targeting this pathway may offer therapeutic potential for treating fungal keratitis caused by Aspergillus fumigatus.

Mesenchymal Stem Cell-Derived Extracellular Vesicles Protect Human Corneal Endothelial Cells from Endoplasmic Reticulum Stress-Mediated Apoptosis

Lola Buono et al.

Summary: The study demonstrated that MSC-derived extracellular vesicles have a significant impact on reducing endoplasmic reticulum stress in corneal endothelial cells, while promoting Akt pathway activation and limiting apoptosis. In contrast, serum-derived extracellular vesicles mainly influenced Akt phosphorylation, with minimal effects on endoplasmic reticulum stress modulation and apoptosis prevention. The effects of MSC-EVs were associated with the transfer of multiple endoplasmic reticulum (ER)-stress targeting miRNAs to corneal endothelial cells.

Mitochondrial ROS Induced Lysosomal Dysfunction and Autophagy Impairment in an Animal Model of Congenital Hereditary Endothelial Dystrophy

Rajalekshmy Shyam et al.

Summary: The Slc4a11 KO mouse model replicates the human disease phenotype associated with CHED, with increased mitochondrial ROS causing edema and endothelial cell loss. We found that although autophagy was activated in the KO mice, there was aberrant autophagy flux and lysosomal dysfunction due to disruption of TFEB signaling by mitochondrial ROS. MitoQ treatment reversed lysosomal dysfunction and improved TFEB nuclear localization, reducing corneal edema and endothelial cell loss in the KO animals.
