The genetics of obesity: from discovery to biology

Ruth J. F. Loos et al.

Summary: In this review, Loos and Yeo summarize our current understanding of the genetic underpinnings of monogenic and polygenic obesity, highlighting shared genetic and biological underpinnings between the two forms of obesity. Genome-wide association studies with increasing sample sizes have driven recent discoveries, with post-GWAS collaborations facilitating translation of genetic loci into meaningful biology and new treatment avenues.

Genome-wide epistasis analysis for Alzheimer's disease and implications for genetic risk prediction

Hui Wang et al.

Summary: This study conducted a genome-wide epistasis analysis to identify novel genetic interactions potentially involved in Alzheimer's disease, and developed a new method to assess the genetic risk of AD. The findings suggest that Epistasis Risk Scores (ERS) can serve as an indicator for genetic risk of AD.

CADD-Splice-improving genome-wide variant effect prediction using deep learning-derived splice scores

Philipp Rentzsch et al.

Summary: The study compared several machine learning methods that score variant effects on splicing using an experimental dataset, integrating the best methods into general variant effect prediction models and evaluating the impact on the classification of known pathogenic variants. The inclusion of splicing DNN effect scores substantially improved predictions across multiple variant categories in the new CADD-Splice model, without compromising overall performance.

Estimation of non-additive genetic variance in human complex traits from a large sample of unrelated individuals

Valentin Hivert et al.

Summary: This study introduces a method to calculate non-additive genetic variance for complex traits by analyzing unrelated individuals with genome-wide SNP data instead of traditional data on relatives. The results show that genetic variance for complex traits is predominantly additive, and large sample sizes are needed to estimate epistatic variance accurately.

Endoplasmic reticulum stress as the basis of obesity and metabolic diseases: focus on adipose tissue, liver, and pancreas

Aline Fernandes-da-Silva et al.

Summary: Obesity challenges lipid and carbohydrate metabolism, leading to glucolipotoxicity and ER dysfunction, triggering ER stress and metabolic abnormalities. Dietary interventions can alleviate ER stress induced by excessive fat or sugar intake, impacting adipose tissue, liver, and pancreas function.

Linking the FTO obesity rs1421085 variant circuitry to cellular, metabolic, and organismal phenotypes in vivo

Samantha Laber et al.

Summary: This study found that variants in FTO are strongly associated with obesity. Through experiments in mice, it was confirmed that the deletion of the conserved cis-regulatory module rs1421085 can affect gene expression and mitochondrial function in adipocytes. The study also showed a cross-species conservation of the regulatory circuitry at molecular, cellular, metabolic, and organismal levels, revealing a previously unknown contextual dependence of the variant's action.
