α-Viniferin and ε-Viniferin Inhibited TGF-β1-Induced Epithelial-Mesenchymal Transition, Migration and Invasion in Lung Cancer Cells through Downregulation of Vimentin Expression

Alpha-viniferin and epsilon-viniferin show significant inhibitory effects on EMT, invasion, and migration in non-small cell lung cancer, and can reverse the expression changes induced by TGF-beta 1.
Resveratrol has well-known anticancer properties; however, its oligomers, including alpha-viniferin, epsilon-viniferin, and kobophenol A, have not yet been well investigated. This is the first study examining the anti-epithelial-mesenchymal transition (EMT) effects of alpha-viniferin and epsilon-viniferin on A549, NCI-H460, NCI-H520, MCF-7, HOS, and U2OS cells. The results showed that alpha-viniferin and epsilon-viniferin significantly inhibited EMT, invasion and migration in TGF-beta 1- or IL-1 beta-induced non-small cell lung cancer. alpha-Viniferin and epsilon-viniferin also reversed TGF-beta 1-induced reactive oxygen species (ROS), MMP2, vimentin, Zeb1, Snail, p-SMAD2, p-SMAD3, and ABCG2 expression in A549 cells. Furthermore, epsilon-viniferin was found to significantly inhibit lung metastasis in A549 cell xenograft metastatic mouse models. In view of these findings, alpha-viniferin and epsilon-viniferin may play an important role in the prevention of EMT and cancer metastasis in lung cancer.
