Increased dosage and treatment time of Epigallocatechin-3-gallate (EGCG) negatively affects skeletal parameters in normal mice and Down syndrome mouse models

Raza Jamal et al.

Summary: This study found that high doses of EGCG treatment in Ts65Dn mice may worsen skeletal structural deficits, indicating the importance of monitoring skeletal phenotypes when administering high doses of EGCG therapeutically.

Design, synthesis and biological evaluation of harmine derivatives as potent GSK-3β/DYRK1A dual inhibitors for the treatment of Alzheimer's disease

Wenwu Liu et al.

Summary: Harmine derivatives were designed, synthesized, and evaluated as dual GSK-3 beta/DYRK1A inhibitors, showing promising activity against both targets. Compound ZDWX-25 exhibited potent inhibitory effects on GSK-3 beta and DYRK1A, with IC50 values of 71 and 103 nM, respectively. These compounds have the potential to be promising candidates for Alzheimer's disease treatment.

Green tea extracts containing epigallocatechin-3-gallate modulate facial development in Down syndrome

John M. Starbuck et al.

Summary: The study found that in the DS mouse model, low doses of GTE-EGCG were beneficial for improving facial skeletal morphology, while early supplementation of EGCG in DS children was associated with reduced facial dysmorphology. However, high doses of GTE-EGCG usage led to increased facial dysmorphology, indicating dose-dependent effects of GTE-EGCG on modulating facial development.

Stress Responses in Down Syndrome Neurodegeneration: State of the Art and Therapeutic Molecules

Chiara Lanzillotta et al.

Summary: Down syndrome is characterized by early onset Alzheimer's disease due to triplication of genes on chromosome 21, leading to oxidative stress and protein oxidation. Therapeutic approaches targeting antioxidant response and protein stability have shown efficacy in murine models, but further studies are needed for clinical translation.

Dyrk1a from Gene Function in Development and Physiology to Dosage Correction across Life Span in Down Syndrome

Helin Atas-Ozcan et al.

Summary: Research has identified Dyrk1a as the key driver gene in Down syndrome, playing a crucial role in brain development and neural function. Targeting DYRK1A overdosage with specific kinase inhibitors shows promising evidence for alleviating cognitive conditions in individuals with Down syndrome. However, addressing the developmental and neurodegenerative aspects of DS across the lifespan remains an open question.

Epigallocatechin-3-Gallate Plus Omega-3 Restores the Mitochondrial Complex I and F0F1-ATP Synthase Activities in PBMCs of Young Children with Down Syndrome: A Pilot Study of Safety and Efficacy

Iris Scala et al.

Summary: The study demonstrates that EGCG plus omega-3 is safe for children with Down syndrome and effective in improving mitochondrial complex I and V activities. However, a decline in plasma folates was observed in some patients treated with EGCG, which requires careful monitoring. Results regarding developmental performance improvements remain inconclusive.
