Molecular signaling in pancreatic ductal metaplasia: emerging biomarkers for detection and intervention of early pancreatic cancer

Loss of Jag1 cooperates with oncogenic Kras to induce pancreatic cystic neoplasms

Summary: Notch signaling in the pancreas has dual roles in cancer, acting both as an oncogene and a tumor suppressor. Deletion of Jag1 in combination with oncogenic Kras(G12D) leads to development of cystic neoplasms rather than ductal adenocarcinoma in mice, with Jag1 acting as a tumor suppressor by delaying precursor lesions. Loss of Jag1 promotes a phenotypic switch from malignant carcinoma to benign cystic lesions. JAG1 expression in pancreatic cancer patients is higher in cancerous tissue, associated with poor overall survival, and correlated with SOX9 expression which also has similar associations with survival outcomes.

Secretory Mucin SAC Promotes Neoplastic Progression by Augmenting KLF4-Mediated Pancreatic Cancer Cell Sternness

Summary: Genetic ablation of secreted mucin 5AC (MUC5AC) in a murine model significantly delays neoplastic onset and PanIN lesion progression in pancreatic cancer, with a reduction in cancer stem cell markers and tumor formation frequency. Mechanistically, MUC5AC potentiates oncogenic signaling through integrin ow135, pSrc (Y416), and pSTAT3 (Y705), leading to enrichment of the self-renewing CSC population. These findings highlight the crucial role of MUC5AC in promoting cancer cell sternness during pancreatic tumorigenesis and suggest it as a potential therapeutic target.
