Comparison of human poly-N-acetyl-lactosamine synthase structure with GT-A fold glycosyltransferases supports a modular assembly of catalytic subsites

Summary: Poly-N-acetyl-lactosamine (poly-LacNAc) structures play a vital role in development, immune function, and human disease. The majority of poly-LacNAc is synthesized by the alternating action of B3GNT2 and galactosyltransferases. The structure of B3GNT2 conserves GT-A fold and DxD motif, supporting a direct displacement inverting catalytic mechanism for UDP-GlcNAc sugar donor.

Structures and mechanism of human glycosyltransferase β1,3-N-acetylglucosaminyltransferase 2 (B3GNT2), an important player in immune homeostasis

Summary: β-1,3-N-acetylglucosaminyltransferases (B3GNTs) are Golgi-resident glycosyltransferases involved in the biosynthesis of poly-N-acetyl-lactosamine chains. Among the characterized B3GNTs, B3GNT2 is the major poly-N-acetyl-lactosamine synthase and its deletion leads to hypersensitive immunocytes. The structural and kinetic studies of human B3GNT2 reveal critical residues and a novel N-terminal helical domain that may stabilize the catalytic domain and distinguish among different acceptor substrates.

FUT8-Directed Core Fucosylation of N-glycans Is Regulated by the Glycan Structure and Protein Environment

Genetic engineering in CHO cells demonstrated that core fucosylation mainly occurs on complex-type N-glycans, but certain glycosites can acquire Fuc on high-mannose N-glycans.
