Deciphering the synergistic and redundant roles of CG and non-CG DNA methylation in plant development and transposable element silencing

Wenjie Liang et al.

Summary: The study revealed that CG and non-CG DNA methylation synergistically and redundantly regulate plant reproductive development, vegetative development, and TE silencing in Arabidopsis. A group of TEs are specifically activated in ddcc met1(+/-) mutants, while size divergence is associated with differences in levels of DNA damage in these mutants.

Roles of DEMETER in regulating DNA methylation in vegetative tissues and pathogen resistance

Wenjie Zeng et al.

Summary: In this study, CRISPR technology was used to edit the DME gene in Arabidopsis, creating weak DME mutants and a quadruple mutant, confirming the important role of DME in DNA demethylation in plant vegetative tissues. The research also revealed that DME, along with three other demethylase genes, play crucial regulatory roles in plant responses to biotic and abiotic stresses.

Genome-wide distribution and functions of the AAE complex in epigenetic regulation in Arabidopsis

Yi-Zhe Zhang et al.

Summary: The AAE complex targets common genomic regions and primarily interacts with genetic heterochromatin on a genome-wide level. It has a significant influence on poly(A) site usage of genes containing heterochromatin, as well as on alternative splicing regulation of overlapping heterochromatin-containing genes like RPP4. Additionally, AAE is involved in epigenetic silencing of transposable elements outside of genes.

The Arabidopsis active demethylase ROS1 cis-regulates defence genes by erasing DNA methylation at promoter-regulatory regions

Thierry Halter et al.

Summary: Active DNA demethylation plays a crucial role in plant and mammalian immunity by promoting basal resistance and erasing methylation at specific promoter regions to facilitate WRKY transcription factors binding, thereby enabling the expression of immune response genes.

Conserved dual-mode gene regulation programs in higher eukaryotes

Jun-Yeong Lee et al.

Summary: Recent studies have revealed a fundamental feature in eukaryotic gene regulation, the dual-mode gene regulation, which is common even in species lacking CGIs. In vertebrates and plants, genes associated with high methylation level promoters are inactivated by heterochromatin formation, while genes with low methylation level promoters are broadly expressed and maintained as euchromatin. In invertebrate animals lacking DNA methylation, some genes are regulated by Polycomb proteins, while others are regulated by heterochromatin formation.

SDC mediates DNA methylation-controlled clock pace by interacting with ZTL in Arabidopsis

Wenwen Tian et al.

Summary: The molecular bases of eukaryotic circadian clocks involve transcriptional-translational feedback loops and epigenetic codes, with DNA methylation playing a significant role in fine-tuning circadian rhythms. Global genome DNA hypomethylation can prolong the circadian period in Arabidopsis, with the SDC protein acting as a link between DNA hypomethylation and a proteolytic cascade to tune the circadian clock.

The mop1 mutation affects the recombination landscape in maize

Meixia Zhao et al.

Summary: The mutation of the MOP1 gene in maize results in decreased meiotic recombination in pericentromeric regions but increased recombination in gene-rich chromosomal arms. The loss of CHH methylation near genes is driving the redistribution of crossovers in mop1 mutants. In contrast, no significant changes were observed in the frequency of meiotic recombination in lbl1 mutants.

Roles of IDM3 and SDJ1/2/3 in Establishment and/or Maintenance of DNA Methylation in Arabidopsis

Wei Miao et al.

Summary: Our analyses show that IDM3 and SDJ1/2/3 are involved in establishing and/or maintaining DNA methylation through the DDR complex, in contrast to previous studies suggesting their role in DNA demethylation pathways. IDm3 and SDJ1/2/3 mutations lead to genome-wide DNA hypomethylation and affect DNA methylation levels and patterns on a genome-wide scale. These findings demonstrate that IDM3 and SDJ1/2/3 play a role in DNA methylation of specific genomic regions through the DDR complex.

Genetic and environmental modulation of transposition shapes the evolutionary potential of Arabidopsis thaliana

Pierre Baduel et al.

Summary: This study reveals that transposable element (TE) mobilization is a major driver of genetic variation in A. thaliana, regulated by genetic and environmental factors. TE insertions occur at a similar rate to base substitutions, with individual TE families showing varying mobilization activities. High rates of transposition seem to be positively selected at the edge of the species' ecological niche.
