Tau-Atrophy Variability Reveals Phenotypic Heterogeneity in Alzheimer's Disease

Sandhitsu R. Das et al.

Summary: The study aimed to investigate the impact of Tau neurofibrillary tangles on neurodegeneration, and identified age and burden of white matter hyperintensity lesions as related modulatory factors. Data analysis showed that grouping individuals based on the relationship between Tau neurofibrillary tangles and neurodegeneration may help identify potential contributors to neurodegeneration.

Data-driven FDG-PET subtypes of Alzheimer's disease-related neurodegeneration

Fedor Levin et al.

Summary: This study explored distinct neurodegeneration subtypes of AD using FDG-PET as a molecular imaging marker. Three main hypometabolic subtypes were identified, showing differential cognitive decline trajectories. These findings may provide a sensitive tool for early detection and characterization of AD-related neurodegeneration variants.

Four distinct trajectories of tau deposition identified in Alzheimer's disease

Jacob W. Vogel et al.

Summary: By using an unbiased subtyping algorithm, the study systematically characterized longitudinal tau variability in human Alzheimer's disease, identifying four trajectories of tau deposition with distinct clinical features. The results suggest that variation in tau pathology is common and systematic, potentially necessitating a re-examination of the notion of 'typical AD' and a revisiting of tau pathological staging.

Astrocyte activation imaging with 11C-acetate and amyloid PET in mild cognitive impairment due to Alzheimer pathology

Michael Tran Duong et al.

Summary: This pilot study evaluated astrocyte activation in the medial temporal lobe in individuals with mild cognitive impairment (MCI) and found that C-11-acetate retention was significantly elevated in MCI compared to cognitively normal participants. The study also showed significant associations between MTL C-11-acetate retention, amyloid burden, and neurocognitive measures in MCI. This suggests a potential role for C-11-acetate PET as a neuroinflammatory biomarker in MCI and early AD.

The development and convergence of co-pathologies in Alzheimer's disease

John L. Robinson et al.

Summary: This study found that CAA, LATE-NC, and Lewy bodies are common pathologies in Alzheimer's disease that worsen in severity as the disease progresses. Age was associated with all pathologies, while the APOE epsilon 4 gene was specifically linked to CAA. In individuals with dementia, age was linked to LATE-NC, while Lewy bodies were more common in younger patients.
