Summary: A series of novel substituted pyrazolo[3,4-c]pyridin-5-ylamidines were synthesized and evaluated for their cytotoxicity against cancer cell lines, showing potential antiproliferative and apoptotic induction capabilities. Certain derivatives exhibited low toxicity against normal cells, along with a G(0)/G(1) arrest of HT-1080 cells. The study also investigated the potential mechanisms of cytotoxic activity, with active compounds inhibiting phosphorylation of Akt and ERK and/or inducing p38 MAPK phosphorylation.
