YTHDF1 Promotes Gastric Carcinogenesis by Controlling Translation of FZD7

Jingnan Pi et al.

Summary: This study reveals the oncogenic role of YTHDF1 in gastric cancer, showing its regulation of the Wnt/β-catenin signaling pathway through m(6)A modification. Inhibition of YTHDF1 can reduce gastric cancer cell proliferation and tumorigenesis, suggesting a new approach for targeting epigenetic regulators in cancer treatment.

Direct Regulation of DNA Repair by E2F and RB in Mammals and Plants: Core Function or Convergent Evolution?

Swarnalatha Manickavinayaham et al.

Summary: RB and E2F proteins collaborate to regulate the cell cycle in eukaryotes and also have important functions in DNA repair. In some organisms, other proteins have taken over the cell cycle regulation function of RB and E2F. Apart from cell cycle regulation, E2F and RB homologs in various organisms also possess non-canonical functions.

The m6A Reader YTHDF1 Facilitates the Tumorigenesis and Metastasis of Gastric Cancer via USP14 Translation in an m6A-Dependent Manner

Xiao-Yu Chen et al.

Summary: The study found that high expression of YTHDF1 in gastric cancer tissues is associated with poor prognosis and acts as an independent prognostic factor for poor survival in patients with GC. YTHDF1 deficiency inhibited cell proliferation and invasion (in vitro), and gastric tumorigenesis and lung metastasis (in vivo), while also inducing cell apoptosis. YTHDF1 promotes USP14 protein translation in an m(6)A-dependent manner.

Recent Advances in Enhancing the Therapeutic Index of PARP Inhibitors in Breast Cancer

Camille Franchet et al.

Summary: PARP inhibitors are used to treat breast cancer with mutated BRCA1/2 HR factors due to their synthetic lethal effect with homologous recombination deficiency. Unfortunately, the high rate of PARP inhibitor resistance in clinical practice has dampened initial hopes. New strategies are being explored to overcome resistance mechanisms and improve the therapeutic index of PARP inhibitors.
