Synergetic Contributions of Viral VP1, VP3, and 3C to Activation of the AKT-AMPK-MAPK-MTOR Signaling Pathway for Seneca Valley Virus-Induced Autophagy

Jiangwei Song et al.

Summary: This study demonstrated that Seneca Valley virus (SVV) infection activates autophagy through multiple signaling pathways, with viral proteins VP1, VP3, and 3C synergistically contributing to the activation of the AKT-AMPK-MAPK-MTOR pathway. These findings provide insight into the finely tuned molecular mechanisms of SVV-induced autophagy and may aid in the development of control strategies against SVV infection.

The PRIDE database resources in 2022: a hub for mass spectrometry-based proteomics evidences

Yasset Perez-Riverol et al.

Summary: PRIDE is the largest data repository of mass spectrometry-based proteomics data in the world, with around 500 datasets submitted per month. In addition to continuous improvements in data pipelines and infrastructure, PRIDE has developed the Spectra Archive and MAGE-TAB file format to enhance sample metadata annotation and access to mass spectra.

Development of a VP2-based real-time fluorescent reverse transcription recombinase-aided amplification assay to rapidly detect Senecavirus A

Wenlong Wang et al.

Summary: In this study, a real-time fluorescent reverse transcription recombinase-aided amplification (rRT-RAA) assay was developed for the detection of Senecavirus A (SVA). The assay showed high sensitivity and specificity, comparable to the previously used PCR assay. Testing results using 189 clinical swine samples demonstrated good consistency and correlation between the rRT-RAA and rRT-PCR assays.

Multi-organ proteomic landscape of COVID-19 autopsies

Xiu Nie et al.

Summary: This study used proteomic analysis to reveal the molecular mechanisms of multi-organ injuries in COVID-19 patients. The data showed significant upregulation of cathepsin L1 in the lung, instead of ACE2. Systemic hyperinflammation, dysregulation of glucose and fatty acid metabolism were detected in multiple organs.

Comparison of historical and contemporary isolates of Senecavirus A

Alexandra C. Buckley et al.

Summary: Senecavirus A (SVA) was discovered in 2002 as a cell culture contaminant, with subsequent reports of porcine idiopathic vesicular disease (PIVD) cases testing PCR positive for SVA. Experimental reproduction of disease with contemporary SVA strains raised questions about reproducing vesicular disease with historical isolates. Comparing historical and contemporary SVA isolates in growing pigs revealed differences in cross-neutralizing antibody levels, but similar clinical presentation of vesicular disease.

Selective autophagy receptor SQSTM1/p62 inhibits Seneca Valley virus replication by targeting viral VP1 and VP3

Wei Wen et al.

Summary: The study explores the reciprocal regulation between the selective autophagy receptor SQSTM1 and Seneca Valley virus (SVV). Overexpression of SQSTM1 decreased viral protein production and reduced viral titers, while SVV evolved strategies to circumvent host autophagic machinery to promote viral replication. SVV's 3C(pro) targets SQSTM1 for cleavage to prevent selective autophagy and promote viral propagation.

Tandem Mass Tag-Based Quantitative Proteomic Analysis of ISG15 Knockout PK15 Cells in Pseudorabies Virus Infection

Wenfeng He et al.

Summary: PRV is a significant pathogen for swine, and the identification of protein targets against PRV is crucial. The study used TMT-based approach to identify 241 differentially expressed proteins in PRV-infected ISG15 knockout cells, potentially playing important roles in PRV propagation. Validation through RT-qPCR and Western blot confirmed the accuracy of TMT results, while bioinformatics analysis revealed involvement in various biological processes and signaling pathways.

Xiangle Zhang et al.

Summary: By comparing the expression features of IBRS-2 and PK-15 cells in response to SVV infection, differences in innate immune response were identified, with aberrant signal transduction found to be the underlying cause of distinct immune response and viral replication rates between the two cell lines.

Comparative Proteomic Profiling: Cellular Metabolisms Are Mainly Affected in Senecavirus A-Inoculated Cells at an Early Stage of Infection

Fuxiao Liu et al.

Summary: This study used proteomics and metabolomics methods to investigate the impact of SVA infection on the proteomic profile of BSR-T7/5 cells. It was found that 361 differentially expressed proteins were identified in SVA-infected cells at 12 hours post-inoculation, with 305 upregulated and 56 downregulated proteins. GO and KEGG enrichment analyses indicated that cellular metabolisms were mainly affected in the early stage of SVA infection.
