Detection and genetic analysis of a novel atypical porcine pestivirus from piglets with congenital tremor in Japan

Miwako Kasahara-Kamiie et al.

Summary: A newly discovered porcine virus, Atypical porcine pestivirus (APPV), associated with congenital tremor (CT) in pigs was identified and its complete genome sequence was analyzed in Japanese piglets with CT. Phylogenetic analysis revealed that the Japanese APPV belonged to genotype 3 and was distantly related to other strains within this genotype. Furthermore, novel genotype 3 APPVs have been prevalent in Japan since at least 2007, as evidenced by retrospective examinations using RT-PCR.

Evolution and genetic diversity of atypical porcine pestivirus (APPV) from piglets with congenital tremor in Guangxi Province, Southern China

Kaichuang Shi et al.

Summary: The study identified a high degree of genetic diversity of atypical porcine pestivirus (APPV) in Guangxi province, Southern China, with distinct differences in nucleotide and amino acid sequences among different strains. Phylogenetic analysis revealed that the APPV strains could be divided into different clades based on gene sequences, and the strains from Guangxi province fell into two separate clades. Evolution analysis indicated an average evolutionary rate and estimated the most recent common ancestor of APPV strains from America, Europe, and Asia.

Genetic characterization of atypical porcine pestivirus (APPV) in China and the successful isolation of a novel APPV strain within genotype 2

Qiaoya Zhang et al.

Summary: The study revealed that Chinese APPV strains showed high conservation in the NS3 gene, while variability was observed in NS5A, Npro, and Erns genes. Chinese APPV strains were classified into multiple genotypes and subgenotypes. Additionally, the successful isolation of a Chinese APPV strain provides a research foundation for future studies.

Fc-Mediated E2-Dimer Subunit Vaccines of Atypical Porcine Pestivirus Induce Efficient Humoral and Cellular Immune Responses in Piglets

Xujiao Ren et al.

Summary: This study aimed to construct APPV E2 subunit vaccines fused with Fc fragments and evaluate their immunogenicity in piglets. The results showed that the Fc fusion proteins emulsified with the ISA 201VG adjuvant elicited stronger humoral and cellular immune responses than the IMS 1313VG adjuvant. The findings suggest that APPV E2 subunit vaccines fused with Fc fragments may be a promising vaccine candidate against APPV.

Summary: Researched prepared VLPs of APPV and evaluated VLPs vaccine in BALB/c mice. Results showed strong antibody responses induced by VLPs, reducing viral load, and BALB/c mice are suitable as a laboratory model for APPV.

MEGA11 Molecular Evolutionary Genetics Analysis Version 11

Koichiro Tamura et al.

Summary: MEGA software, a mature tool for computational molecular evolution, has been enhanced with new additions for building timetrees of species, pathogens, and gene families using rapid relaxed-clock methods. These additions include methods for estimating divergence times, confidence intervals, and new options for tagging sequences with spatiotemporal sampling information. The enhancements aim to improve user experience, result quality, and the pace of biological discovery.

Disruption of interferon-β production by the Npro of atypical porcine pestivirus

Chunxiao Mou et al.

Summary: The study revealed that APPV-N-pro is capable of inhibiting IFN-beta production by interacting with IRF3 and interfering with its phosphorylation, primarily blocking IRF3 activity to combat IFN-beta. Additionally, it was found that the N-terminal 31-51 amino acids of APPV-N-pro were mainly associated with inhibiting the IFN-beta response.
