A New Insight into the Potential Role of Tryptophan-Derived AhR Ligands in Skin Physiological and Pathological Processes

ZFP57 dictates allelic expression switch of target imprinted genes

β-naphthoflavone-induced upregulation of CYP1B1 expression is mediated by the preferential binding of aryl hydrocarbon receptor to unmethylated xenobiotic responsive elements

Summary: The study found that DNA methylation inhibits CYP1B1 expression by interfering with the binding of AhR to target sequences. It also showed that pre-treatment of HepG2 cells with 5-aza-2'-deoxycytidine enhanced the induction of CYP1B1 expression by beta-naphthoflavone, suggesting the positive effect of demethylation on XRE2/XRE3 sequences. The research provides the first direct evidence that ligand-activated AhR preferentially binds to unmethylated XRE sequences in natural chromatin, and the approach can be applied to study the effects of DNA methylation on target sequence binding by transcription factors other than AhR.

Individual DNA Methylation Pattern Shifts in Nanoparticles-Exposed Workers Analyzed in Four Consecutive Years

Summary: This study analyzed the effect of chronic exposure to nanoparticles on DNA methylation patterns in individuals over four consecutive years. The results showed shifts in DNA methylation patterns between years in both exposed and control subjects, with greater differences observed in the exposed group over a three-year period. Specific cg loci showed relatively stable methylation changes in chronically exposed individuals. Overall, long-term exposure to nanoparticles can lead to fixed epigenetic changes related to specific environmental factors, such as inhalation of nanoparticles.
