The pyruvate-lactate axis modulates cardiac hypertrophy and heart failure

Summary: The metabolic rewiring of cardiomyocytes, including decreased mitochondrial pyruvate oxidation and increased lactate export, is a key feature of heart failure. The mitochondrial pyruvate carrier (MPC) and the cellular lactate exporter monocarboxylate transporter 4 (MCT4) play pivotal roles in this metabolic axis. Alteration of the pyruvate-lactate axis is a fundamental and early feature of cardiac hypertrophy and failure.

Uranium inhibits mammalian mitochondrial cytochrome c oxidase and ATP synthase

Summary: Uranium as an emerging pollutant presents serious threats to ecological and human health, with the kidney identified as a major deposition site and sensitive target organ for its toxicity, which is associated with oxidative stress and mitochondrial respiration. This study found that cytochrome c oxidase and ATP synthase play key roles in uranium-induced toxicity by inhibiting their activities in the mitochondrial respiration pathway. These inhibitory effects could explain the generation of reactive oxygen species and mitochondrial alterations observed in uranium poisoning.

Alterations in the gut microbiota and metabolite profiles in the context of neuropathic pain

Summary: The study revealed significant differences in gut microbiota composition and metabolite profiles between rats with CCI-induced neuropathic pain and sham controls, with specific changes in abundance of certain genera. Additionally, 72 significantly differentially expressed serum metabolites and 17 significantly differentially expressed spinal cord metabolites were identified between the CCI rats and the sham rats, highlighting the potential role of gut microbiota and related metabolites in modulating metabolic disturbances in neuropathic pain.

Tryptophan Pathway-Targeted Metabolomics Study on the Mechanism and Intervention of Cisplatin-Induced Acute Kidney Injury in Rats

Summary: The study revealed that tryptophan metabolism was significantly disturbed in the rat kidney medulla and cortex after cisplatin administration, with the medulla being more sensitive. Indoxyl sulfate, accumulating significantly in a dose-dependent manner, was identified for further study. Chlormethiazole, a CYP2E1 inhibitor, was found to reduce the production of indoxyl sulfate in the liver and attenuate cisplatin-induced AKI in rats.
