MMP2 and MMP9 Activity Is Crucial for Adult Visual Cortex Plasticity in Healthy and Stroke-Affected Mice

Ipek Akol et al.

Summary: The extracellular matrix (ECM) plays a crucial role in neuronal network development and plasticity. Proteolytic cleavage of ECM and cell surface proteins is important for regulating plasticity. MMP2 and MMP9 are enzymes involved in proteolysis and their activity is critical for adult visual plasticity.

Reduced Severity of Tissue Injury Within the Infarct May Partially Mediate the Benefit of Reperfusion in Ischemic Stroke

Felix C. Ng et al.

Summary: This study found that the severity of tissue injury within the infarct, as determined by assessing the integrity of the blood-brain barrier (BBB), is independently associated with improved functional outcome in patients with ischemic stroke. In addition to reducing infarct volume, reperfusion may also improve clinical outcome by reducing tissue injury severity within the infarct.

Impact of aging and comorbidities on ischemic stroke outcomes in preclinical animal models: A translational perspective

Eduardo Candelario-Jalil et al.

Summary: Ischemic stroke is a complex and devastating neurological disease, with neuroprotective strategies targeting penumbral tissue playing a crucial role in minimizing brain damage. Current reperfusion therapies have limitations, leading to high interest in combining them with neuroprotective strategies. Preclinical models of ischemic stroke involving aged animals and comorbidities are critical for successful drug development and translation to clinical trials.

Blood-Brain Barrier Disruption and Hemorrhagic Transformation in Acute Ischemic Stroke: Systematic Review and Meta-Analysis

Francesco Arba et al.

Summary: The systematic review and meta-analysis confirmed the association between BBB disruption and hemorrhagic transformation in patients with acute ischemic stroke. CT studies showed more uniform application and consistent results compared to MR studies, indicating the need for standardization of protocols and methodology for implementing BBB disruption as a neuroradiological marker in clinical practice.

Interleukin-10 improves stroke outcome by controlling the detrimental Interleukin-17A response

Marius Piepke et al.

Summary: This study investigated the role of IL-10 in ischemic stroke and found that IL-10 plays a key function in controlling IL-17A signaling. IL-10 acts differently on alpha beta and gamma delta T cells, and it was also discovered that IL-17A could be a therapeutic opportunity for stroke treatment.

Molecular Magnetic Resonance Imaging of Vascular Inflammation After Recanalization in a Rat Ischemic Stroke Model

Bart A. A. Franx et al.

Summary: Molecular MRI can detect vascular inflammation after acute ischemic stroke, providing complementary information for treatment decision-making. In the study, it was found that VCAM-1 expression significantly increased at 6 hours post-recanalization and declined after 24 hours, but remained elevated in lesion borderzones.

TSPO imaging in animal models of brain diseases

Nadja Van Camp et al.

Summary: TSPO-PET imaging has been considered as the gold standard biomarker reference for measuring increased neuroinflammation, with easier interpretation in acute focal injury models and more challenges in chronic models with lower or diffuse microglial activation. Despite some technical limitations, TSPO imaging remains a valuable tool for studying the time course or response to treatment of neuroinflammation in both acute and chronic disease models.

Chronic T cell proliferation in brains after stroke could interfere with the efficacy of immunotherapies

Steffanie Heindl et al.

Summary: Stroke is a major cause of death and permanent disability globally, with current therapies limited to vascular recanalization in the acute phase. Neuroinflammation has emerged as a key focus in translational research, playing a crucial role in post-stroke recovery and neurodegeneration.

Revisiting the neurovascular unit

Samantha Schaeffer et al.

Summary: Schaeffer and Iadecola reviewed the heterogeneity of the neurovasculature and discussed the limitations of the neurovascular unit concept and the molecular diversity of cerebrovascular cells. They proposed the concept of a 'neurovascular complex' composed of segmentally diverse functional modules that implement coordinated vascular responses to central and peripheral signals.

Multimodal Imaging with NanoGd Reveals Spatiotemporal Features of Neuroinflammation after Experimental Stroke

Violaine Hubert et al.

Summary: This study proposes and validates a preclinical in vivo MRI tool to monitor neuroinflammation following ischemic stroke, based on injection of a novel multimodal nanoprobe designed for internalization by phagocytic cells. The study confirms efficient internalization of NanoGd by microglia and shows hypointense signals in the ischemic lesion using in vivo MRI with NanoGd injection in a mouse model. Additionally, the study demonstrates NanoGd internalization by activated phagocytic cells through both intravital microscopy and ex vivo analysis.

In vivo multimodal imaging of adenosine Al receptors in neuroinflammation after experimental stroke

Ana Joya et al.

Summary: The study demonstrates the overexpression of A(l)ARs in microglia and infiltrated leukocytes after cerebral ischemia, with pharmacological modulation showing a decrease in inflammation and improvement in neurological outcome. Activation of A(l)AR led to reduction of brain lesion and improvement of motor, sensory, and reflex responses, highlighting the key role of A(l)AR in controlling glial activation and proliferation in response to stroke.
