PillarX: A Microfluidic Device to Profile Circulating Tumor Cell Clusters Based on Geometry, Deformability, and Epithelial State

A new insight to deformability correlation of circulating tumor cells with metastatic behavior by application of a new deformability-based microfluidic chip

Summary: The study developed a novel continuous microfluidic platform for capturing and characterizing circulating tumor cells (CTCs) based on their deformability differences. The deformability-based chip demonstrated high efficiency and purity in isolating cancer cells, as well as accurate categorization based on deformability differences. Analysis of clinical blood samples from breast cancer patients showed that the chip was able to detect CTCs, process high throughput blood samples, and identify CTCs with potential for metastasis based on deformability differences.

Mechanical Adaptability of Tumor Cells in Metastasis

Summary: The most dangerous aspect of cancer is its potential for metastasis, as tumor cells must undergo a series of biomechanical steps to form life-threatening metastases in distant organs. Successful cancer cells may need specific cell mechanical properties, including high deformability, to adapt to the changing microenvironmental constraints along their journey. The mechanical signature of a successful cancer cell may be its ability to dynamically refine its properties according to the challenges encountered at each step of the metastatic cascade.

Detection of clustered circulating tumour cells in early breast cancer
