Dysregulation of the orexin/hypocretin system is not limited to narcolepsy but has far-reaching implications for neurological disorders

Chiara Berteotti et al.

Summary: Neuropeptides orexin A and B are selectively released by neurons in the hypothalamus, playing a key role in regulating wake-sleep state switching and other physiological functions. Dysfunction of the orexin system has been associated with narcolepsy, Alzheimer's disease, stroke, and Prader-Willi syndrome, highlighting the importance of orexin system integrity for overall health.

Noradrenergic circuit control of non-REM sleep substates

Alejandro Osorio-Forero et al.

Summary: During NREMS, fluctuation of noradrenaline levels in thalamus are associated with sleep spindle rhythms and heart-rate variations, indicating vigilance-promoting mechanism by LC neurons. Optogenetic activation or inhibition of LC neurons can modulate sleep-spindle clustering and heart-rate variations, affecting sensory arousability during NREMS. Noradrenergic modulation of thalamic circuits plays a key role in maintaining sensory arousability in mammalian NREMS.

MRI-assessed locus coeruleus integrity is heritable and associated with multiple cognitive domains, mild cognitive impairment, and daytime dysfunction

Jeremy A. Elman et al.

Summary: The study found that reduced LC integrity is associated with widespread differences across cognitive domains, daytime sleep-related dysfunction, and increased risk for aMCI. These findings highlight the potential of MRI-based measures of LC integrity in early identification of AD risk in late-middle-aged adults.

Tau-driven degeneration of sleep- and wake-regulating neurons in Alzheimer's disease

Caroline H. Lew et al.

Summary: Disturbances of the sleep/wake cycle in Alzheimer's disease are common and tend to worsen with disease progression. These disturbances are likely driven by abnormal tau protein accumulation in sleep/wake regulating areas, emphasizing the importance of early diagnosis and treatment for sleep issues in AD patients.

Summary: This review highlights the important role of noradrenergic projections from the locus coeruleus in regulating cognition and behavior, as well as their loss in neurodegenerative diseases. The connections of Locus Coeruleus with hypothalamus: potential involvement in Alzheimer's disease

Filippo Sean Giorgi et al.

Summary: The hypothalamus and Locus Coeruleus play important roles in regulating the sleep/wake cycle and autonomic activities, with complex anatomical connections linking the two structures. In Alzheimer's disease, disruptions in the hypothalamic network and degeneration of the Locus Coeruleus may contribute to pathophysiological processes. The impairment of Locus Coeruleus may partly explain the occurrence of hypothalamic-related symptoms in AD.

Late-in-life neurodegeneration after chronic sleep loss in young adult mice

Summary: Short-term sleep loss impacts A beta and tau levels, while sleep disturbances predict cognitive decline. Chronic short sleep exposure leads to late-life neurodegeneration and disrupts amyloid and tau homeostasis.

Summary: Individuals show varied responses to stressful events, with research findings indicating that fMRI-measured responsivity of the human brainstem arousal system and associated pupil responses are related to the severity of psychopathological symptoms triggered by prolonged real-life stress.

In vivo and neuropathology data support locus coeruleus integrity as indicator of Alzheimer's disease pathology and cognitive decline

Heidi I. L. Jacobs et al.

Summary: Various autopsy studies have shown that the locus coeruleus (LC) is the initial site of hyperphosphorylated TAU aggregation, and as TAU accumulates in LC neurons, pathology spreads throughout the cortex. Lower LC integrity is associated with increased TAU deposition in the entorhinal cortex in older individuals, and LC pigmentation ratings correlate with LC neuronal density, particularly in advanced Braak stages. These findings suggest that LC integrity may be a promising indicator of early Alzheimer's disease processes and cognitive changes.

Noradrenaline in the aging brain: Promoting cognitive reserve or accelerating Alzheimer's disease?

Mara Mather

Summary: Engaging in novel and mentally challenging activities may promote brain health and prevent Alzheimer's disease, but could also come with risks as more active brain regions accumulate more pathology over time. Noradrenaline released during such activities may have protective effects, but can also accelerate the production and accumulation of amyloid-beta and tau in the brain.

Summary: The study found abnormal LC-NE functional network in patients with CID, and the altered LC-NE function in dACC was associated with anxiety symptoms in CID. This extends the understanding of the neuropathological basis of CID and provides a potential treatment target for CID patients with anxiety.
