Evaluation of the carcinogenicity of dichloromethane in rats, mice, hamsters and humans

Wolfgang Dekant et al.

Summary: Dichloromethane (DCM) is classified as a cat. 2 carcinogen, mainly inducing malignant tumors in mice but not in rats and hamsters, with human data not supporting a conclusion of cancer risk. The dose-dependent toxicokinetics of DCM suggest it is a threshold carcinogen in mice, while rats and hamsters have low activities of the DCM-metabolizing enzyme GSTT1 which is more relevant in humans. The current classification of DCM as a cat. 2 human carcinogen remains appropriate based on species-specific toxicokinetics and tumor induction in mice.
