Acquired temozolomide resistance in MGMTlow gliomas is associated with regulation of homologous recombination repair by ROCK2

Summary: This study found that high expression of ROCK2 is correlated with poor overall survival in MGMT(low) recurrent gliomas. Overactive ROCK2 enhances homologous recombination repair in TMZ-resistant glioma cell lines with low MGMT expression, while silencing ROCK2 impairs repair and induces cell death. These findings suggest the potential use of ROCK inhibitors in clinical therapy for MGMT(low) TMZ-resistant gliomas.

Inhibition of HSP90 as a Strategy to Radiosensitize Glioblastoma: Targeting the DNA Damage Response and Beyond

Summary: The study highlights the potential of HSP90 inhibitors as radiosensitizers in glioblastoma treatment. Inhibition of HSP90 led to decreased DNA damage response factors, altered transcriptomics, impaired DNA damage repair, and reduced clonogenic survival in response to radiation in vitro. While in vivo results were promising in terms of tumor progression and survival outcomes, further optimization is required to enhance the efficacy of the combination therapy.
