CNS genomic profiling in the mouse chronic social stress model implicates a novel category of candidate genes integrating affective pathogenesis

Konstantin A. Demin et al.

Summary: Despite high prevalence, medical impact and societal burden, anxiety, depression and other affective disorders remain poorly understood and treated. Clinical complexity and polygenic nature complicate their analyses. Experimental findings show that mouse affective phenotypes exhibit complex temporal dynamics as they develop.

Transcriptomic organization of the human brain in post-traumatic stress disorder

Matthew J. Girgenti et al.

Summary: This study provides comprehensive insights into the molecular pathology of post-traumatic stress disorder (PTSD) through transcriptome analysis of postmortem brains, revealing extensive remodeling of gene expression networks and identifying genetic susceptibility factors, as well as marked transcriptional sexual dimorphism. Additionally, comparison with major depressive disorder cohorts shows significant differences between the molecular profiles of individuals with PTSD and major depressive disorder despite their high comorbidity, supporting the notion that PTSD has distinct molecular determinants.

Glucocorticoids, metabolism and brain activity

Aneta Jaszczyk et al.

Summary: The review integrates various experimental approaches to better understand the effects of elevated glucocorticoids on brain activity and metabolism. It suggests that glucocorticoids alter neuronal activity dynamics, leading to context-specific changes in both excitation and inhibition, which support task-related responses. Glucocorticoids also diversify energy sources for the brain and impact carbohydrate reserves, with potential stress due to mismatches in supply and utilization.

Kv1.1 channels mediate network excitability and feed-forward inhibition in local amygdala circuits

Samrat Thouta et al.

Summary: The study identified the crucial roles of K(v)1.1 subunits in controlling excitability in the amygdala, with a specific focus on BLA pyramidal neurons and CeL neurons. Loss of K(v)1.1 resulted in enhanced intrinsic excitability in CeL neurons and impaired inhibitory synaptic transmission, leading to dysfunction of feed-forward inhibition. This study highlights the importance of inhibitory control in preventing recurrent hyperexcitation related to seizures in Kcna1(-/-) mice.

Curation of over 10 000 transcriptomic studies to enable data reuse

Nathaniel Lim et al.

Summary: Despite the vast amount of transcriptomic data in public repositories, effective reuse remains challenging due to issues such as unstructured metadata, inconsistent data processing, and varying probe-gene mappings. The Gemma bioinformatics system was created to address these issues by providing curated transcriptomic datasets, analytical software, and a web interface. With over 10,000 manually curated datasets and hundreds of curated transcriptomic platforms, Gemma offers broad coverage of conditions and tissues, particularly focusing on brain-related datasets. Users can access the curated data and differential expression analyses through the Gemma website, RESTful service, and an R package.
