Fluorescence Correlation Spectroscopy Monitors the Fate of Degradable Nanocarriers in the Blood Stream

Sascha Schmitt et al.

Summary: The use of nanoparticles as carriers to deliver pharmacologically active compounds to specific parts of the body via the bloodstream is a promising therapeutic approach. However, monitoring and identifying the nanoparticle carriers and their cargo in the blood environment is challenging. Researchers have developed a new method based on fluorescence correlation spectroscopy that allows accurate measurement of the size, concentration, and loading efficiency of the nanoparticles, and tracking the fate of fluorescently labeled nanogels in the blood of live mice.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Squaric Ester-Based, pH-Degradable Nanogels: Modular Nanocarriers for Safe, Systemic Administration of Toll-like Receptor 7/8 Agonistic Immune Modulators

Anne Huppertsberg et al.

Summary: pH-responsive nanogels were fabricated for safe delivery of TLR7/8-stimulating imidazoquinolines, showing a spatially controlled immunostimulatory activity in the spleen after intravenous injection, without causing systemic off-target inflammatory responses. This approach highlights the potential of pH-degradable nanogels as a promising platform for intravenous administration of small-molecular TLR7/8 agonists in immune therapeutic applications.

Targeting macrophages in cancer immunotherapy

Zhaojun Duan et al.

Summary: Immunotherapy is considered the most promising treatment for cancers, with various strategies such as adoptive cellular immunotherapy, tumor vaccines, antibodies, immune checkpoint inhibitors, and small-molecule inhibitors showing success. The review highlights the role of macrophages in current immunotherapies and the potential benefits of targeting them, suggesting that therapeutic strategies involving macrophages have the potential to enhance the efficacy of cancer treatments.
