Understanding the role of cysteine in ferroptosis: progress & paradoxes

Carson D. Poltorack et al.

Summary: Cysteine is conditionally essential for protein synthesis and intracellular metabolites, and its depletion can trigger nonapoptotic cell death. Despite being normally maintained at low levels within cells, mechanisms to buffer cysteine depletion seem ineffective in cancer cells. The regulation of cysteine and its contribution to ferroptosis are discussed in this article.

Cancers associated with human gammaherpesviruses

Kwun Wah Wen et al.

Summary: EBV and KSHV, both human gammaherpesviruses with oncogenic properties, are associated with various lymphoproliferative diseases and plasmacytic/plasmablastic neoplasms. These viruses can also infect epithelial cells and cause carcinomas. The pathogenesis of these two viruses is closely linked to the expression of viral proteins.

Lipid Metabolism in Ferroptosis

Zhi Lin et al.

Summary: Lipid metabolism plays a crucial role in regulating cell survival and death. Abnormal lipid metabolism can lead to ferroptosis, an iron-dependent regulated cell death. Various antioxidant systems and membrane repair pathways help mitigate oxidative damage and support cell survival, especially in pathological conditions like cancer. Impairment of the ferroptosis machinery is associated with the development of diseases such as cancer and tissue damage.

Epstein-Barr Virus-Encoded Latent Membrane Protein 1 and B-Cell Growth Transformation Induce Lipogenesis through Fatty Acid Synthase

Michael Hulse et al.

Summary: The study demonstrates that ectopic expression of LMP1 and EBV-mediated B-cell growth transformation induces significant changes in fatty acids and lipid droplet formation. Inhibiting lipogenesis leads to preferential killing of LMP1-expressing B cells and hinders EBV immortalization of primary B cells. Targeting LMP-induced lipogenesis could be an effective strategy in treating LMP1-positive EBV-associated malignancies, distinguishing their treatment from EBV-negative counterparts.

DHODH-mediated ferroptosis defence is a targetable vulnerability in cancer

Chao Mao et al.

Summary: The study reveals the mechanism of ferroptosis induced by inhibiting GPX4 in cancer cells, as well as the potential pathways to enhance or inhibit ferroptosis by intervening in relevant metabolic pathways. Therapeutic strategies targeting different levels of GPX4 expression could be a promising new approach for cancer treatment.

Epstein-Barr Virus Induced Cytidine Metabolism Roles in Transformed B-Cell Growth and Survival

Jin Hua Liang et al.

Summary: EBV infection is associated with various cancers, with CTPS1 and CTPS2 potentially playing important roles in EBV-infected B cells. Lack of CTPS1 can impact EBV DNA synthesis, while deficiency of both CTPS1 and CTPS2 leads to stronger phenotypes, suggesting CTPS1/2 as potential therapeutic targets for EBV-driven lymphoproliferative disorders.

Multifaceted mechanisms mediating cystine starvation-induced ferroptosis

Zhennan Shi et al.

Summary: Recent studies have shown additional mechanisms behind cystine starvation-induced ferroptosis, apart from impaired GSH synthesis, highlighting the importance of the cyst(e)ine/glutathione/glutathione peroxidase 4 (GPX4) axis in triggering ferroptosis and inhibiting tumor growth.
