B Cells and Tertiary Lymphoid Structures: Friends or Foes in Cancer Immunotherapy?

Martin Lauss et al.

Summary: Tumor-associated B cells and tertiary lymphoid structures (TLS) play important roles in cancer immunology. B cells have various functions such as antigen presentation, antibody production, and regulation. TLSs in tumors show substantial heterogeneity and support immune response. Current research is exploring the role of TLSs in immunotherapy, and results indicate that TLSs are beneficial for patient prognosis.

Regulatory B cells in cancer

Daniel Michaud et al.

Summary: Tumorigenesis involves genetic mutations and changes in the microenvironment, with immune cells in tumors playing a role in promoting tumor growth. Recent studies have shed light on the mechanisms of how B cells regulate immune responses, with negative regulation potentially impacting anti-tumor immune function through cytokine production.

B cells and cancer: To B or not to B?

Wolf Herman Fridman et al.

Summary: Recent studies have suggested that B cells in the tumor microenvironment may play a significant role in presenting tumor-associated antigens to T cells and producing antibodies that enhance antigen presentation, ultimately impacting cancer clinical outcomes. Immune complexes formed by B cells can also have deleterious effects through inflammation, angiogenesis, and immunosuppression via macrophage and complement activation.

Single-Cell Transcriptomic Analysis Reveals a Tumor-Reactive T Cell Signature Associated With Clinical Outcome and Immunotherapy Response In Melanoma

Min Yan et al.

Summary: The infiltration of tumor-reactive T cells in the tumor site is associated with better survival and immunotherapy response. Through single-cell RNA sequencing analysis, a tumor-reactive-related signature (TRS) was identified and validated to evaluate the tumor reactivity of T cells in melanoma patients. High TRS scores correlated with strong immune activity, high mutation burden, and better survival outcomes, with predictive potential for immunotherapy response.

Single-Cell Transcriptomic Analyses Define Distinct Peripheral B Cell Subsets and Discrete Development Pathways

Alexander Stewart et al.

Summary: The separation of B cells into different subsets has provided insights into their diverse functions in immune responses. Advances in single-cell techniques have enabled a deeper understanding of B cell characteristics and functions, though challenges remain. This study offers a reference single-cell dataset for better classification of functional B cell subsets in peripheral blood, highlighting the complexity and diversity of B cell populations.

Role of Tumor-Infiltrating B Cells in Clinical Outcome of Patients with Melanoma Treated With Dabrafenib Plus Trametinib

Jan C. Brase et al.

Summary: Although patients with unresectable or metastatic melanoma can experience long-term survival with targeted therapies, resistance is common and pretherapeutic biomarkers are lacking. A study on previously untreated BRAF V600-mutant melanoma patients treated with dabrafenib plus trametinib found that baseline cell-cycle gene expression signature and T-cell/B-cell gene signatures were associated with clinical outcomes, highlighting the potential predictive role of B cells in treatment response. Further validation in larger patient cohorts is needed to confirm these findings.

Summary: Understanding intratumoral heterogeneity through single-cell analyses has revealed new biomarkers, but lacks information on cellular location within the tumor microenvironment. New technologies enabling detection of cancer subclones in their native spatial context promise to drive the next generation of cancer research, diagnosis, and therapeutic strategies.

CD86+ Antigen-Presenting B Cells Are Increased in Cancer, Localize in Tertiary Lymphoid Structures, and Induce Specific T-cell Responses

Kerstin Wennhold et al.

Summary: The study found that BAPCs were increased in the tumor microenvironment of various cancer types, especially in patients with colorectal cancer and gastroesophageal adenocarcinoma. BAPCs were localized in lymphoid follicles of TLS, inducing autologous T-cell responses, which provides a mechanistic rationale for the correlation between B-cell abundance and response to immune checkpoint inhibition.
