New ingenane diterpenoids from Euphorbia kansui reverse multi-drug resistance

Summary: Ten ingenane-type diterpenoids, including two newly described compounds, were isolated from the roots of Euphorbia kansui. These compounds were found to be potent chemoreversal agents with low cytotoxicity against adriamycin-resistant human breast adenocarcinoma cells. In particular, compounds 1-2 exhibited greater chemoreversal ability than the positive drug verapamil.

Lathyrane Diterpenoids as Novel hPXR Agonists: Isolation, Structural Modification, and Structure-Activity Relationships

Summary: The study identified a series of potent hPXR agonists from a library of lathyrane diterpenoids, showing better efficacy than the typical hPXR agonist rifampicin. The most active compound 8 could dose-dependently activate hPXR and up-regulate the expressions of PXR downstream genes. These findings suggest that lathyrane diterpenoids could be a new type of hPXR agonist for future anticholestasis drug development.
