The Devastating Clinical Consequences of Child Abuse and Neglect: Increased Disease Vulnerability and Poor Treatment Response in Mood Disorders

Elizabeth T. C. Lippard et al.

Summary: A large body of evidence has demonstrated the long-term consequences of childhood maltreatment, which increases the risk for psychiatric and medical disorders. This review summarizes the effects of childhood maltreatment on vulnerability to mood disorders, highlighting the increased prevalence and associated risks. It discusses the persistent alterations in the hypothalamic-pituitary-adrenal axis and inflammatory cytokines, as well as candidate genes, environmental factors, and neurobiological associations that may mediate these effects. The studies provide insights into modifiable mechanisms and guide treatment and prevention strategies.

Genome-wide association study of patients with a severe major depressive episode treated with electroconvulsive therapy

Caitlin C. Clements et al.

Summary: Analysis of severe cases of major depressive disorder (MDD) using genome-wide SNP genotyping revealed a higher burden of genetic risk loci, stronger genetic correlations with other psychiatric disorders, and weaker correlations with personality and stress-related traits, suggesting a different genetic architecture at the severe end of the MDD spectrum.

Combined effects of genotype and childhood adversity shape variability of DNA methylation across age

Darina Czamara et al.

Summary: Studies on the impact of childhood adversity on DNA methylation (DNAm) are inconsistent, with joint effects of genotype and prenatal environment providing the best explanation for variability in DNAm at birth. In adults, interactions between genotype and childhood adversity show the strongest contribution to DNAm variability.

The interaction of child abuse and rs1360780 of the FKBP5 gene is associated with amygdala resting-state functional connectivity in young adults

Christiane Wesarg et al.

Summary: Extensive research has shown that the rs1360780 SNP in the FKBP5 gene interacts with early-life stress to predict psychopathology. Individuals with the TT genotype of rs1360780 who experienced child abuse exhibit differences in brain structure and function related to emotion processing. An interaction effect was observed between rs1360780 and child abuse in predicting resting-state functional connectivity, suggesting that the TT genotype may increase vulnerability to functional changes related to emotions and bodily sensations in individuals with a history of abuse.

Highly accurate protein structure prediction with AlphaFold

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

The differential calibration of the HPA axis as a function of trauma versus adversity: A systematic review and p-curve meta-analyses

Niki Hosseini-Kamkar et al.

Summary: There is conflicting evidence on whether early-life adversity leads to increased or decreased cortisol reactivity. P-curve analysis revealed that trauma is associated with blunted cortisol reactivity, while adversity is linked to heightened cortisol reactivity. This suggests an inverted U-shaped relationship between severity of adversity and cortisol reactivity.

GxEsum: a novel approach to estimate the phenotypic variance explained by genome-wide GxE interaction based on GWAS summary statistics for biobank-scale data

Jisu Shin et al.

Summary: The study introduces a new method called GxEsum for estimating the phenotypic variance explained by genotype-environment interaction on a genome-wide scale. Through comprehensive simulations and analysis on a large dataset from the UK Biobank, it was found that GxEsum has high computational efficiency and can handle large-scale biobank datasets.

Rethinking Concepts and Categories for Understanding the Neurodevelopmental Effects of Childhood Adversity

Karen E. Smith et al.

Summary: Understanding the processes through which early adverse experiences affect children's nervous-system development, health, and behavior is crucial for developing effective interventions. However, current conceptualizations are limited, and alternative formulations are needed to advance knowledge about the neurobiological mechanisms through which adversity influences human development.
