Inflammatory Bowel Diseases (IBD) and the Microbiome-Searching the Crime Scene for Clues

Mirae Lee et al.

Summary: Inflammatory bowel diseases (IBD) result from the interaction of environmental, microbial, immunological, and genetic factors, with alterations in the gut microbiota playing a role in their development and progression. The use of microbiome-based therapies can potentially prevent, manage, and cure IBD, bridging the gap between basic sciences and clinical application through research findings.

Berberine improves colitis by triggering AhR activation by microbial tryptophan catabolites

Wanghui Jing et al.

Summary: The study showed that berberine can effectively improve the pathological phenotype, gut barrier disruption, and colon inflammation in rats with DSS-induced colitis. Furthermore, berberine can alleviate gut dysbiosis in rats and improve gut barrier function through the regulation of microbial metabolites to activate AhR.

Qingchang Huashi Formula attenuates DSS-induced colitis in mice by restoring gut microbiota-metabolism homeostasis and goblet cell function

Jingyi Hu et al.

Summary: The study aimed to define the effect of QHF and its components, Baitouweng (PBR) and Baizhi (ADR) on treating UC. Results showed that QHF significantly inhibited colitis and protected mice from weight loss and colon shortening. ADR and PBR were found to have strong efficacy in inhibiting DSS-induced colitis.

Role of the IL23/IL17 Pathway in Crohn's Disease

Heike Schmitt et al.

Summary: Crohn's disease is a chronic inflammatory disorder of the gastrointestinal tract influenced by genetic susceptibility, environmental factors, and intestinal microbiome. Pro-inflammatory cytokines IL17 and IL23 play crucial roles in the pathogenesis of CD by causing excessive activation of the mucosal immune system. Treatments targeting these cytokines, such as selective IL23 inhibitors, have shown promising results in CD patients who failed previous anti-TNF therapy.

Gut microbiota-derived inosine from dietary barley leaf supplementation attenuates colitis through PPARγ signaling activation

Daotong Li et al.

Summary: Our study demonstrates that the gut microbiota-inosine-A(2A)R/PPAR gamma axis is crucial in maintaining intestinal homeostasis and may offer a novel approach for preventing colitis through manipulation of gut microbial purine metabolite. Additionally, dietary supplementation of barley leaf improves intestinal mucosal barrier functions and induces metabolic reprogramming in colonic tissues by enhancing the glycolysis process. Furthermore, the enrichment of microbiota-derived purine metabolite inosine from BL supplementation activates PPAR gamma signaling in human colon epithelial cells.

Current Strategies and Potential Prospects of Nanomedicine-Mediated Therapy in Inflammatory Bowel Disease

Fengqian Chen et al.

Summary: Inflammatory bowel diseases (IBD) such as Crohn's disease and ulcerative colitis are highly debilitating due to the imbalance of inflammatory mediators within the inflamed bowel. Conventional drugs for IBD treatment suffer from bioavailability issues and systemic side effects, while nanoparticle (NP)-derived therapy has shown improvements in therapeutic efficacy and targeting specificity. Recent studies have highlighted the fast-growing field of nanomedicines tailored to the pathophysiology of bowel diseases, offering potential for prolonged circulation period and reduced side effects through improved drug encapsulation and targeted delivery.
