Human guanylate binding proteins: nanomachines orchestrating host defense

Miriam Kutsch et al.

Summary: Disease-causing microorganisms breach anatomical barriers and invade tissues, entering host cells rich in nutrients, where host cell-autonomous defense programs are crucial. Dynamin-related guanylate binding proteins (GBPs) play a central role in human cell-autonomous immunity, with recent studies providing a molecular understanding of how GBP nanomachines execute immunity. These studies have revealed GBP1 as a pattern recognition receptor for bacterial lipopolysaccharide, linking GMP production by GBP1 to inflammasome activation, and identifying GBP2 and GBP5 as inhibitors of viral envelope glycoprotein processing, improving our conceptual understanding of GBP-mediated immunity and guiding future research.

Functions of ROS in Macrophages and Antimicrobial Immunity

Marc Herb et al.

Summary: Reactive oxygen species (ROS) play essential roles in various cellular processes, including antimicrobial activity and immune signaling regulation. However, in-depth studies on ROS and their regulated target molecules are still limited. This review provides an overview of ROS sources, functions, and related probes, scavengers, and inhibitors.

Overexpression screen of interferon-stimulated genes identifies RARRES3 as a restrictor of Toxoplasma gondii infection

Nicholas Rinkenberger et al.

Summary: The study investigated the role of IFN gamma-induced ISGs in restricting Toxoplasma gondii infection, and identified RARRES3 as a novel factor restricting parasite infection. The limited number of ISGs capable of restricting T. gondii infection when individually expressed suggests that IFN gamma-mediated immunity to T. gondii infection is a complex, multifactorial process.

Mitochondrial Reactive Oxygen Species Regulate Immune Responses of Macrophages to Aspergillus fumigatus

Remi Hatinguais et al.

Summary: This study highlights the crucial role of mitochondrial ROS (mitoROS) in macrophage antifungal responses, specifically in the regulation of cytokine secretion through RET-mediated mitoROS production. Furthermore, NOX2 is demonstrated to be essential for regulating cytokine secretion by RET-associated mitoROS in response to infection with swollen conidia.

Summary: Human xanthine oxidoreductase (XOR) is a versatile enzyme with various activities, including purine catabolism, production of reactive oxygen species, generation of nitric oxide, and NADH oxidase activity. These activities contribute to metabolism of compounds and play a key role in inflammatory defense.
