Deconstructive Asymmetric Total Synthesis of Morphine-Family Alkaloid (-)-Thebainone A

Summary: The development of a deconstructive strategy for the first asymmetric synthesis of (-)-thebainone A was described, utilizing an enantioselective C-C bond activation and a C-O bond cleavage reaction. The newly optimized conditions show broad substrate scope and excellent enantioselectivity (up to 99.5:0.5 er). The synthesis of (-)-thebainone A was completed by taking advantage of boron-mediated ether bond cleavage through two complementary routes.

Intermolecular [5+2] Annulation between 1-Indanones and Internal Alkynes by Rhodium-Catalyzed C-C Activation

Summary: The study presents a [5+2] cycloaddition reaction catalyzed by Rh, which activates C-C bonds to enable the cyclization of 1-indanones and internal alkynes. The reaction demonstrates tolerance towards various functional groups and provides a direct route to diverse benzocycloheptenones. DFT studies suggest that migration insertion is the turnover-limiting step, with beneficial pi-pi interactions in the transition states.

Site-Selective C-C Cleavage of Benzocyclobutenones Enabled by a Blocking Strategy Using Nickel Catalysis

Summary: In this study, an unprecedented highly selective cleavage of C1-C8 bond with the insertion of alkynes was achieved by using a blocking strategy via Ni catalysis. This provides an efficient method for the synthesis of 1,8-disubstituted naphthalenes, with the potential for easy removal of the blocking group after the transformation.

Skeleton Reorganization of Substituted Benzocyclobutenols through Rh-Catalyzed C-C Bond Cleavage Manipulated by Hydrogen Transfer

Summary: This study unveiled a skeleton reorganization of alkene-tethered benzocyclobutenols through Rh-catalyzed C-C bond cleavage coupled with intra- and intermolecular hydrogen transfer, leading to the synthesis of divergent benzofurans bearing 4-beta-hydroxy or 4-beta-keto moieties under pH- and redox-neutral conditions. The reaction pathway was finely tuned by the catalytic systems.

Summary: A Pd-catalyzed chemo-, regio-, and enantioselective ring-closing/ring-opening cross coupling reaction has been developed with diverse aryl halide-tethered alkenes and benzocyclobutenols as substrates. This method provides a convenient approach to chiral 2,3-dihydrobenzofurans with excellent enantioselectivities, and has been applied in the concise synthesis of analogues of cannabinoid receptor 2 agonists.
