Safety and Feasibility of Nucleus Accumbens Surgery for Drug Addiction: A Systematic Review

Paula Alejandra Navarro et al.

Summary: This study conducted a systematic review of 15 studies on NAcc surgery for substance dependence treatment, finding that while available studies report benefits in treating drug addictions with NAcc surgery, further carefully planned prospective studies are needed to address the efficacy and indications.

Cocaine increases stimulation-evoked serotonin efflux in the nucleus accumbens

Jason Yuen et al.

Summary: Although dopamine is the most implicated neurotransmitter in addiction, serotonin also plays a vital role. This study used N-shaped fast-scan cyclic voltammetry to investigate the acute effects of cocaine on phasic serotonin release in the nucleus accumbens core. The findings contribute to our understanding of the mechanism of action of cocaine and provide a baseline for further research in cocaine addiction.

Biomarkers for Deep Brain Stimulation in Animal Models of Depression

Jason Yuen et al.

Summary: Despite recent advances in depression treatment, many patients still do not respond to serial conventional therapies and are considered treatment resistant. Deep brain stimulation (DBS) has therapeutic potential in this context. This comprehensive review of recent studies of DBS for depression in animal models identifies potential biomarkers for improving therapeutic efficacy and predictability of conventional DBS to aid future development of closed-loop control of DBS system.

Therapeutic deep brain stimulation disrupts movement-related subthalamic nucleus activity in parkinsonian mice

Jonathan S. Schor et al.

Summary: This study investigated the therapeutic effects and underlying mechanisms of subthalamic nucleus deep brain stimulation (STN DBS) in Parkinson's disease mice. The results suggest that STN DBS exerts its therapeutic effects through disrupting movement-related activity in the subthalamic nucleus. These findings provide new insights for optimizing Parkinson's disease treatments and offer an approach for studying deep brain stimulation in other neuropsychiatric conditions.

Tonic Serotonin Measurements In Vivo Using N-Shaped Multiple Cyclic Square Wave Voltammetry

Hojin Shin et al.

Summary: N-MCSWV is a novel voltammetric technique that allows quantitative measurements of extracellular levels of serotonin and other substances in vivo. When used with PEDOT:Nafion-coated carbon fiber microelectrodes, it can continuously monitor tonic serotonin concentrations in the brain of rats.

Consolidating the Circuit Model for Addiction

Christian Luscher et al.

Summary: Addiction is a disease characterized by compulsive drug seeking and consumption, with altered synaptic transmission identified as the functional substrate of pathological behavior. Recent research has focused on integrating circuits underlying compulsion and negative reinforcement, providing blueprints for novel addiction therapies. Genetic and epigenetic correlates of individual vulnerability in addiction are also being explored, with a gain-of-function explanation for circuit remodeling emerging as a key aspect.

Error-Related Activity in Striatal Local Field Potentials and Medial Frontal Cortex: Evidence From Patients With Severe Opioid Abuse Disorder

Elena Sildatke et al.

Summary: The study aimed to investigate the involvement of the nucleus accumbens in performance monitoring, using simultaneous recordings of EEG and LFP. Significant modulations in LFPs related to accuracy were observed during error processing and subjective error awareness, demonstrating the functional interrelation of striatal and cortical processes in performance monitoring.

Population-specific neuromodulation prolongs therapeutic benefits of deep brain stimulation

Teresa A. Spix et al.

Summary: This study demonstrates that manipulating distinct neural subpopulations in the GPe through optogenetics can provide long-lasting therapeutic effects in dopamine-depleted mice, surpassing the effects of traditional DBS. By using synaptic differences and brief bursts of electrical stimulation to excite and inhibit different types of GPe neurons, the circuit-inspired DBS produced sustained therapeutic benefits extending beyond conventional DBS.

Summary: Deep brain stimulation (DBS) is a powerful neurostimulation therapy for neuropsychiatric disorders, with its effects on brain dynamics and reversibility depending on the selected target. Stimulation of the lateral hypothalamus (LH) shows long-lasting effects on brain metabolism and neuroplasticity.

Subthalamic low-frequency oscillations predict vulnerability to cocaine addiction

Mickael Degoulet et al.

Summary: Identifying vulnerable individuals before they transition to compulsive drug seeking is a challenge in addiction prevention. Abnormal oscillatory activity in the STN predicts compulsive-like behavior, and STN DBS at 30 Hz reduces pathological cocaine seeking in compulsive individuals. Early electrical signatures of future compulsive behaviors can inform addiction treatment strategies.

Dopamine 'ups and downs' in addiction revisited

Anne-Noel Samaha et al.

Summary: Repeated drug use can alter dopamine function, leading to addiction. Two main viewpoints exist: one suggests that drug use decreases dopamine neurotransmission, while the other argues that it enhances it. Recent evidence supports the latter view, with implications for addiction modeling and treatment.

Synaptic mechanism underlying serotonin modulation of transition to cocaine addiction

Yue Li et al.

Summary: This study found that cocaine use enhances compulsive behavior by affecting serotonin levels, while this effect can be reversed by increasing serotonin levels. The presynaptic depression induced by 5-HT via 5-HT1B receptors plays a role in the bidirectional effect on behavior.

Summary: For over 40 years, in vivo microdialysis techniques have been crucial in studying the effects of illicit substances on dopamine levels in the brain, but limitations have led to the development of a new electrochemical method known as M-CSWV. This method provides faster recordings of dopamine level changes, allowing for a more detailed understanding of neuronal responses to drug administration.

Summary: This study evaluated the safety of DBS for substance use disorder and its effects on substance use, craving, emotional symptoms, and frontal/executive functions. The results showed that DBS of the NAc/VC was safe and effective in reducing substance use and craving, as well as improving frontal and executive functions. Confirmation of these findings with future studies is needed.
