Dissecting the Shared Genetic Architecture of Suicide Attempt, Psychiatric Disorders, and Known Risk Factors

Niamh Mullins et al.

Summary: A genome-wide association study identified significant genetic loci associated with suicide attempts, particularly one locus on chromosome 7 which is implicated in risk-taking behavior, smoking, and insomnia. The study also revealed strong genetic correlations between suicide attempts and psychiatric disorders, as well as other nonpsychiatric traits like smoking, pain, and lower socioeconomic status. Conditioning on psychiatric disorders reduced the genetic correlations between suicide attempts and psychiatric disorders, indicating a shared biology with nonpsychiatric risk factors.

Mendelian randomisation for psychiatry: how does it work, and what can it tell us?

Robyn E. Wootton et al.

Summary: The successful prevention of mental illness relies upon the identification of causal, modifiable risk factors. Mendelian randomisation (MR) is a method that uses genetic variation to identify causal effects between risk factors and outcomes in a cost-effective and time-efficient manner. While MR studies are increasing in psychiatry, there are limitations to consider, along with the importance of clear causal questions and sensitivity analyses.

BRAINMECHANISMS OF INSOMNIA: NEW PERSPECTIVES ON CAUSES AND CONSEQUENCES

Eus J. W. Van Someren

Summary: Insomnia is the second most common mental disorder, yet progress in understanding its underlying neurobiological mechanisms has been limited. This review explores the definition, prevalence, and susceptibility factors of insomnia, including genetic variants, early life stress, major life events, and brain structure and function. The proposed model suggests that in individuals vulnerable to developing insomnia, the locus coeruleus may be more sensitive to salience network input, leading to a spiral of hyperarousal and impaired sleep.

Summary: The study explored the relationships between sleep health and various clinical phenotypes, finding associations with circulatory system, mental disorders, endocrine/metabolic phenotypes, obesity, major depressive disorder, hypercholesterolemia, and acne. The study supports the use of PheWAS as a promising tool for advancing sleep research.

Insomnia in the context of short sleep increases suicide risk

Hedstrom Anna Karin et al.

Summary: This study found that insomnia is only associated with an increased suicide risk among individuals with short sleep duration, with no significant association observed among those who slept 7 hours or more. The total effect of insomnia in the context of short sleep on suicide risk was 2.85, with a direct effect of 2.25 and an indirect effect mediated by depression of 1.27. 32% of the total effect was mediated by depression, and the association between insomnia and suicide risk was more pronounced with decreasing depressive symptoms.

The relationship between nightmares, depression and suicide

Anna Karin Hedstrom et al.

Summary: The study found no association between nightmares and suicide among participants with depression, but those with nightmares had a higher risk of developing depression during follow-up. Therefore, interventions targeting both depression and nightmares may provide additional therapeutic benefit.

The heritability of insomnia: Systematic review and meta-analysis of twin studies

Juan J. Madrid-Valero et al.

Summary: Twin studies consistently show that genetic factors play a significant role in explaining variance for insomnia, with a mean heritability estimate of 0.39. However, there is considerable heterogeneity among studies, highlighting the need for further research to identify variables that may explain this variation.

We need to do better: A systematic review and meta-analysis of diagnostic test accuracy of restless legs syndrome screening instruments

Summary: This study evaluated the diagnostic accuracy of screening instruments for RLS and found that no screening instruments can be recommended for use without an expert clinical interview in epidemiological studies. Future instruments should focus on maximizing specificity to determine true prevalence. The study provides guidelines for RLS ascertainment in epidemiological studies and includes an RLS reporting checklist due to the poor reporting quality of many RLS epidemiological studies.

Genetic determinants of daytime napping and effects on cardiometabolic health

Hassan S. Dashti et al.

Summary: Daytime napping is a common heritable behavior, with potential causal links to cardiometabolic health. Genome-wide association studies have identified multiple loci associated with sleep and obesity-related genes, indicating a complex genetic basis for daytime napping and its impact on health outcomes.

Insomnia, Suicidal Ideation, and Suicide Attempts in the Clinical Antipsychotic Trials of Intervention Effectiveness

Brian J. Miller et al.

Summary: Insomnia is associated with suicidal ideation, recent suicide attempt, and greater psychopathology in schizophrenia. Terminal insomnia is significantly associated with current suicidal ideation, while initial/middle insomnia is significantly associated with a recent suicide attempt. Insomnia appears to be an important indicator in the clinical care of patients with schizophrenia.
