[11C]MODAG-001-towards a PET tracer targeting α-synuclein aggregates

Summary: The newly developed MODAG-001 PET tracer based on anle138b showed high affinity in vitro and good penetration and stability in the mouse brain. However, no detectable binding to aggregated alpha SYN was found in human brain sections of LBD cases, possibly due to low abundance of aggregated alpha SYN against background protein.

Pinpointing Brain TREM2 Levels in Two Mouse Models of Alzheimer's Disease

Summary: This study demonstrated the potential of using TREM2 as an in vivo biomarker for microglial activation in Alzheimer's disease. By designing a bispecific antibody capable of crossing the blood-brain barrier, changes in TREM2 levels could be monitored using PET imaging. Transgenic animals showed higher PET radioligand exposure and TREM2 protein levels compared to wild-type mice.

Age-related increase of alpha-synuclein oligomers is associated with motor disturbances in L61 transgenic mice

Summary: The study found an age progressive increase of alpha-synuclein oligomers in the brain of L61 transgenic mice, which were associated with more severe motor symptoms. Male mice displayed increased brain levels of oligomers at an earlier age and a more severe behavioral phenotype compared to females, showing a gender difference.

Summary: This study compared the brain pharmacokinetics of two BBB-penetrating bispecific antibodies of different sizes and found that the smaller non-IgG antibody di-scFv3D6-8D3 exhibited faster elimination from blood, lower brain C-max, higher parenchymal-to-capillary concentration ratio, and earlier brain elimination compared to the larger IgG-based bispecific antibody mAb3D6-scFv8D3. The study also indicated that di-scFv3D6-8D3 had lower avidity towards TfR1, potentially contributing to its faster brain delivery.
