Direct Comparison of the Tau PET Tracers 18F-Flortaucipir and 18F-MK-6240 in Human Subjects

Alexandra Gogola et al.

Summary: This study compared the characteristics of two frequently used tau PET tracers, F-18-flortaucipir and F-18-MK-6240, in the same subjects. The results showed that both tracers performed equally well in visual interpretations and were capable of quantifying signal in a common set of brain regions that develop tau pathology in Alzheimer disease. However, they also showed distinct patterns of apparent off-target binding, and F-18-MK-6240 exhibited a greater dynamic range in SUVR estimates.

Summary: Alzheimer's disease is projected to see a significant increase in prevalence in the future, with genetics playing a key role. Novel biomarkers and lifestyle-based prevention trials show promising therapeutic potential. Promising pharmacological treatments are advancing in clinical trials, targeting amyloid beta, tau, and inflammation.

Reversible Tau Phosphorylation Induced by Synthetic Torpor in the Spinal Cord of the Rat

Timna Hitrec et al.

Summary: The study aimed to investigate the reversible accumulation of PP-Tau in the dorsal horns of the spinal cord during synthetic torpor. ATP-IF was prominently expressed in the dorsal horns during torpor, disappearing after 6 hours of returning to normal temperature. Tau-IF levels also decreased during torpor but returned to normal during recovery. Additionally, microglia activation was not observed in the spinal cord during synthetic torpor.

Discriminatory ability of next-generation tau PET tracers for Alzheimer's disease

Steven Y. Yap et al.

Summary: A new generation of tau PET tracers has been developed for the imaging of Alzheimer's disease and other dementias with high specificity, showing potential to differentiate Alzheimer's disease from other dementias in clinical applications. Despite some remaining challenges, these new tracers have the potential to play an important role in clinical practice.

A current view on Tau protein phosphorylation in Alzheimer's disease

Summary: The functions of Tau in the central nervous system are regulated by various posttranslational modifications, with phosphorylation being widely recognized as a pathological hallmark in diseases like AD. However, the similarities and heterogeneity of phosphorylation patterns between physiological and pathological Tau make it challenging to identify specific modifications as therapeutic targets and biomarkers.
