Brain ventricles as windows into brain development and disease

Phan Q. Duy et al.

Summary: Dilation of fluid-filled cerebral ventricles, known as ventriculomegaly, is a common feature of hydrocephalus and is often seen in autism and schizophrenia. Recent research indicates that studying the genomics of congenital hydrocephalus may provide valuable insights into neural stem cell regulation, human cerebrocortical development, and the pathogenesis of neuropsychiatric diseases.

Parallel in vivo analysis of large-effect autism genes implicates cortical neurogenesis and estrogen in risk and resilience

Helen Rankin Willsey et al.

Summary: Gene Ontology analyses have repeatedly identified synaptic function and transcriptional regulation as key points of convergence in autism spectrum disorders (ASD) risk genes. Leveraging Xenopus tropicalis, a study of ten genes associated with ASD found that mutations led to an increase in neural progenitor cells, implicating cortical neurons in ASD vulnerability and expanding the range of convergent functions to include neurogenesis. Systematic chemical screening identified estrogen, through Sonic hedgehog signaling, as a resilience factor that may mitigate a range of ASD genetic risks in Xenopus and human brain models.

Sequencing of 53,831 diverse genomes from the NHLBI TOPMed Program

Daniel Taliun et al.

Summary: The TOPMed program aims to study the genetic architecture and biology of heart, lung, blood, and sleep disorders to improve diagnosis, treatment, and prevention of these diseases. Resources include a variant browser, genotype imputation server, and genomic and phenotypic data available through dbGaP. The study detected a large number of rare genetic variants, providing insights into mutation processes and recent human evolutionary history.

PTEN mutations in autism spectrum disorder and congenital hydrocephalus: developmental pleiotropy and therapeutic targets

Tyrone DeSpenza et al.

Summary: Research suggests that there is shared molecular pathophysiology between autism spectrum disorder (ASD) and congenital hydrocephalus (CH), with the PTEN-PI3K-mTOR pathway potentially serving as a common underlying mechanism for these two conditions. This provides diagnostic, prognostic, and therapeutic promise for individuals with ASD and CH.
