A DNA Nanostructure-Based Neuroprotectant against Neuronal Apoptosis via Inhibiting Toll-like Receptor 2 Signaling Pathway in Acute Ischemic Stroke

Mi Zhou et al.

Summary: The study demonstrated that tFNA effectively prevented neuronal apoptosis in ischemic stroke by interfering with ischemic cascades in vitro and ameliorating the microenvironment of the ischemic hemisphere in vivo. This pleiotropic nanoneuroprotectant shows promise as a therapeutic strategy for ischemic stroke by attenuating neurological deficits and reducing infarction volume.

Facilitating In Situ Tumor Imaging with a Tetrahedral DNA Framework-Enhanced Hybridization Chain Reaction Probe

Bowen Zhang et al.

Summary: A DNA-enhanced HCR detection system (T-probe system) is introduced for the detection of cancer-related targets, demonstrating its versatility and efficiency by detecting miRNA-21 and nucleolin. The T-probe system shows high detection rate and rapid recognition of both membrane and intracellular cancer-related targets, and exhibits superior fluorescence for imaging tumor-related biomarkers in vitro and in vivo, indicating its great potential for clinical diagnosis and therapeutic applications.

Biomimetic Nanoerythrosome-Coated Aptamer-DNA Tetrahedron/Maytansine Conjugates: pH-Responsive and Targeted Cytotoxicity for HER2-Positive Breast Cancer

Wenjuan Ma et al.

Summary: DNA materials have been used as nanocarriers for targeted cancer therapy, but their short half-life and low bioavailability hinder their clinical translation. This study developed a HER2-targeted DNA-aptamer-modified DNA tetrahedron/DM1 conjugate, and applied a biomimetic camouflage to enhance drug delivery and tumor-stimulated drug release. The hybrid erythrosome-based nanoparticles exhibited superior inhibition of HER2-positive cancer.

Functionalizing Framework Nucleic-Acid-Based Nanostructures for Biomedical Application

Tao Zhang et al.

Summary: This article discusses the strategies for functionalizing diverse tetrahedral framework nucleic acids (tFNAs) and their potential biomedical applications. tFNAs have the ability to scavenge reactive oxygen species, enhance cellular endocytosis and tissue permeability, and promote cell-material interactions. They can be used to treat inflammatory and degenerative diseases, and also have potential applications in targeted therapies, tissue regeneration, antitumor strategies, and antibacterial treatment through their structural programmability.

Anti-inflammatory activity of curcumin-loaded tetrahedral framework nucleic acids on acute gouty arthritis

Mei Zhang et al.

Summary: Gouty arthritis is a common inflammatory arthritis, and controlling inflammation is crucial for prevention. Scientists have synthesized a new material called Cur-TFNAs to enhance the effectiveness and bioavailability of curcumin, a natural anti-inflammatory drug. Cur-TFNAs showed better anti-inflammatory effects in in vivo and in vitro experiments compared with free curcumin.
