C/EBPβ sustains the oncogenic program of AML cells by cooperating with MYB and co-activator p300 in a transcriptional module

Summary: Transcription factor MYB is an important regulator of gene expression in hematopoietic cells and a potential drug target for acute myeloid leukemia (AML). A natural compound, helenalin acetate (HA), has been found to inhibit AML cell viability and induce cell death and differentiation by disrupting the MYB-induced gene expression program. Interestingly, CCAAT-box/enhancer binding protein beta (C/EBP beta), a transcription factor known to cooperate with MYB and the co-activator p300 in myeloid cells, rather than MYB itself, was identified as the primary target of HA. This highlights a novel role for C/EBP beta as a proleukemogenic factor.

A synthetic covalent ligand of the C/EBPβ transactivation domain inhibits acute myeloid leukemia cells

Summary: C/EBP β is a transcription factor that cooperates with MYB to maintain proliferation and differentiation block of AML cells. A synthetic analog of helenalin inhibits C/EBP β by covalently binding to cysteine residues in the transactivation domain, leading to enhanced differentiation, cell death, and reduced self-renewal potential of AML cells. Gene expression profiling confirms the relevance of C/EBP β as a therapeutic target. However, further development is needed to improve the effectiveness of the synthetic analog as a C/EBP β inhibitor.

Recent detection of MYB gene rearrangements and mutations in human cancer provides smoking gun evidence of the involvement of MYB family members in human malignancies. Studies linking the activity of MYB family members to human cancers and experimental therapeutic interventions tailored for MYB-expressing cancers are highlighted in this review.
