Antimalarial application of quinones: A recent update

Om P. S. Patel et al.

Summary: Atovaquone belongs to a class of drugs known as naphthoquinones and is used in combination with proguanil to treat acute, uncomplicated malaria caused by Plasmodium falciparum. Various quinone-derived compounds have been studied for their antimalarial properties, with a recent focus on the discovery of novel quinone-based compounds between 2009 and 2020. The compilation of these discoveries is expected to contribute to the development of effective and safe treatments against malaria.

Summary: The design and synthesis of new 2-phenyl(pyridinyl)benzimidazolequinones and their 5-phenoxy derivatives demonstrate potent anti-Trypanosoma cruzi activity. Compound 11a shows significantly higher activity compared to nifurtimox. Molecular docking studies suggest that these compounds could have a multitarget profile.

Novel plastoquinone analogs containing benzocaine and its analogs: structure-based design, synthesis, and structural characterization

Nilufer Bayrak et al.

Summary: Introducing new biologically active organic molecules or their analogs into pharmaceutically important molecules significantly enhances the discovery and development of medicines. Chemical synthesis plays a key role in pharmaceutical research and development by designing and constructing molecules with essential biological activity. In this study, novel PQ analogs were designed and synthesized from commercially available dimethylhydroquinone, and their antibacterial and antifungal activities were evaluated.

Summary: Cholinesterases are important targets for regulating cholinergic neurotransmission, with 1,4-benzoquinone derivatives showing potential as both antioxidants and inhibitors in neurological disorders. This study designed and synthesized novel 1,4-benzoquinone analogues, evaluated their selective inhibitory potential on AChE and BChE, and conducted behavioral and biochemical studies to understand their effects. The results provided valuable insights into the structural features needed for designing highly selective and potent molecules for cognitive decline management.

Plastoquinone analogs: a potential antimicrobial lead structure intensely suppressing Staphylococcus epidermidis and Candida albicans growth

Emel Mataraci-Kara et al.

Summary: This study evaluated the antimicrobial activity of 25 Plastoquinone analogs, identifying PQ1 as the most potent against Staphylococcus epidermidis and PQ1, PQ2, and PQ7 as the best antifungal agents against Candida albicans. Further investigation on PQ1 and PQ25 showed they can reduce biofilm mass, endorsing efforts to develop them into potential antimicrobial drug candidates.
