Targeting Insulin Resistance to Treat Cognitive Dysfunction

Anit Tyagi et al.

Summary: Dementia, particularly Alzheimer's disease and vascular dementia, is strongly associated with aging. Recent studies have highlighted the potential link between type 2 diabetes mellitus, cognitive dysfunction, and the development of Alzheimer's disease and other cognitive disorders. Research is increasingly focusing on the physiological functions of insulin in the brain, the evaluation of cognitive function under conditions of insulin resistance, and the potential use of antidiabetic drugs to treat dementia.

Catalpol Weakens Depressive-like Behavior in Mice with Streptozotocin-induced Hyperglycemia via PI3K/AKT/Nrf2/HO-1 Signaling Pathway

Xiaohui Wu et al.

Summary: Catalpol has antidepressant effects on depressive behaviors in hyperglycemic state, potentially attributed to upregulation of the PI3K/AKT/Nrf2/HO-1 signaling pathway to restore oxidative-antioxidant balance. This provides preclinical experimental evidence for the application of catalpol in treating depression associated with hyperglycemia.

Ultrasound-assisted extraction conditions optimisation using response surface methodology from Mitragyna speciosa (Korth.) Havil leaves

Fazila Zakaria et al.

Summary: The optimisation of the Ultrasound-Assisted Extraction method was achieved using Response Surface Methodology, resulting in an extraction yield and Total Phenolic Content of 22.69% and 143.51 mg gallic acid equivalent (GAE)/g, respectively. The Liquid Chromatography-tandem Mass Spectrometry analysis identified major phytochemicals in the M. speciosa extract, including catechin, rutin, kaempferol, and more, which could exhibit therapeutic effects such as anti-inflammatory and antioxidant properties.

Swertiamarin mitigates nephropathy in high-fat diet/streptozotocin-induced diabetic rats by inhibiting the formation of advanced glycation end products

Kirti Parwani et al.

Summary: The study investigated the potential of Swertiamarin (SM) in preventing diabetic nephropathy (DN) induced by advanced glycation end products (AGEs) in high fat diet-streptozotocin-induced diabetic rats. The results showed that SM could ameliorate DN by inhibiting oxidative stress induced by AGEs and preventing their binding to the receptor for advanced glycation end products (RAGE), suggesting a promising therapeutic effect in diabetes-related kidney complications.

Isoliquiritigenin attenuates acute renal injury through suppressing oxidative stress, fibrosis and JAK2/STAT3 pathway in streptozotocin-induced diabetic rats

Leiming Sun et al.

Summary: The study evaluated the protective effects and mechanisms of isoliquiritigenin on acute renal injury. Isoliquiritigenin effectively ameliorated cellular viability, reduced ROS content, and upregulated antioxidant enzymes in renal cells. Long-term administration of isoliquiritigenin improved diabetic characteristics, reduced oxidative stress, and inhibited fibrosis- and inflammation-related factors in DN rats through JAK2/STAT3 signaling pathway inhibition.

Myrsinoic acid B from Myrsine coriacea reverses depressive-like behavior and brain oxidative stress in streptozotocin-diabetic rats

Priscila Laiz Zimath et al.

Summary: The study found that treatment with MAA or MAB could prolong the first immobility latency in diabetic rats, while HEBMC administration reduced immobility time and increased climbing in the FST. However, only MAB treatment could shorten immobility time, increase climbing and swimming in FST, and increase the crossing of diabetic animals in the OFT. This behavioral improvement was accompanied by a reduction in oxidative stress in the HIP and PFC.

Accelerated Solvent Extractions (ASE) of Mitragyna speciosa Korth. (Kratom) Leaves: Evaluation of Its Cytotoxicity and Antinociceptive Activity

Yong Sean Goh et al.

Summary: The study utilized the green accelerated solvent extraction technique to produce a better, non-toxic, and antinociceptive active botanical extract of Mitragyna speciosa (kratom). The extract had a short extraction time and required a reduced amount of solvents. It contained a substantial amount of total phenolic and flavonoids, and was found to be non-cytotoxic to HEK-293 and HeLa Chang liver cells. In mice, the extract showed a better antinociceptive effect compared to other extracts, supporting its potential as a safe and cost-effective choice for further preclinical studies.

Role of Quercetin in Depressive-Like Behaviors: Findings from Animal Models

Serena Silvestro et al.

Summary: Depressive-like behavior is a prevalent neuropsychiatric disorder with complex pathophysiology, and current pharmacotherapy is often ineffective and associated with adverse effects. Phytochemical compound quercetin, found in fruits and vegetables, has gained interest for its antioxidant properties and potential antidepressant effects through regulation of genes and pathways targeted by drugs. Preclinical studies in the last decade have shown that quercetin pre-treatment can mitigate depressive-like behaviors by its antioxidant, anti-inflammatory, and neuroprotective effects, suggesting its potential as a nutraceutical or adjunct therapy against stress-induced depressive behavior.
