Summary: A series of cinnamide compounds linked with 1-aryl piperazines, derived from cinnamic acid derivatives, were synthesized and evaluated for their inhibitory activity against mushroom tyrosinase. The presence of a 3-chloro-4-fluorophenyl moiety at the N-1 position of the piperazine ring was found to be essential for a potent inhibitory effect. Molecular docking studies were conducted to investigate the interactions between the compounds and the active site of the enzyme. Some of the highly active compounds also showed potential in inhibiting melanogenesis. However, certain compounds exhibited toxicity at high concentrations.

Improved yield of theaflavin-3,3′-digallate from Bacillus megaterium tyrosinase via directed evolution

Summary: Theaflavin-3,3'-digallate (TFDG) from black tea has various health benefits, but its application is limited due to low yields. In this study, tyrosinases from Bacillus megaterium were used to synthesize TFDG through directed evolution and high-throughput screening. Mutant Bmtyrc-3 showed significantly improved catalytic efficiency and higher production of TFDG. The broader substrate binding area of Bmtyrc-3 resulted in increased activity of substrates EGCG and ECG.

Improving tyrosinase inhibitory activity of grass carp fish scale gelatin hydrolysate by gastrointestinal digestion: Purification, identification and action mechanism

Summary: Biologically active peptides with tyrosinase inhibitory activity from grass carp fish scale gelatin hydrolysates have been obtained and tested for their efficacy in whitening foods. The digested products showed stronger tyrosinase inhibitory activity compared to the original hydrolysates. Six peptides with tyrosinase inhibitory activity were identified using bioaffinity ultrafiltration, with FTGML showing the highest inhibitory activity. The interaction between FTGML and tyrosinase was mainly driven by hydrogen bonding and hydrophobic interaction. In vivo studies using zebrafish models confirmed the whitening and lightning effects of FTGML at specific concentrations.

Clinical trials on the beneficial effect of collagen peptides on skin are also discussed. The prospects and challenges of collagen peptides are emphasized in this review. Collagen peptides have the potential to be an effective ingredient in the food and cosmetics industry with wide application prospects.
