Human germline nuclear transfer to overcome mitochondrial disease and failed fertilization after ICSI

Maoxing Tang et al.

Summary: This study aims to explore the application of human nuclear transfer (NT) in treating mitochondrial disease transmission and failed fertilization. The experiments confirmed the effectiveness of NT in reducing mitochondrial transmission and demonstrated for the first time that NT can enable successful fertilization in cases of failed fertilization.

Mitochondria: emerging therapeutic strategies for oocyte rescue

Zhixin Jiang et al.

Summary: Mitochondria play a crucial role in oocyte development and reproduction, with abnormalities in quantity, quality, and function being associated with decreased fertility and various disorders. Research on using autologous mitochondrial transfer as a therapeutic approach for infertility shows promise, but currently faces technical and clinical challenges that need to be addressed.

Maternal spindle transfer for mitochondrial disease: lessons to be learnt before extending the method to other conditions?

Charalampos Siristatidis et al.

Summary: Mitochondrial diseases are genetic conditions caused by mutations in genes that regulate mitochondrial function. Maternal spindle transfer (MST) has been proposed as a method to prevent the transmission of these diseases, with a successful case reported in Mexico in 2017. However, concerns have been raised about research governance and criticisms associated with the expansion of this technique.

Effects of intracytoplasmic sperm injection timing and fertilization methods on the development of bovine spindle transferred embryos

Yu-Ju Lee et al.

Summary: This study aimed to develop a novel procedure for producing bovine cytoplasmic transfer (CT) embryos, and the results showed that performing intracytoplasmic sperm injection (ICSI) before CT could improve the embryonic development of CT bovine embryos. Moreover, using in vitro fertilization (IVF) instead of ICSI for CT oocyte fertilization dramatically increased the pregnancy rate and birth rate of CT calves.

Oocyte quality and aging

Ali Reza Eftekhari Moghadam et al.

Summary: It is well known that female reproductive ability decreases in the fourth decade of life due to age-related changes in oocyte quality and quantity. However, the causes and physiological aspects of reproductive aging are still unclear. This review paper aims to study the potential impact of the aging process on oocyte quality and female reproductive ability, and presents evidence showing a decrease in oocyte quality with age, resulting in reduced reproductive outcomes for women.

Summary: Somatic cell nuclear transfer (SCNT) is a beneficial tool for aiding the conservation of endangered species, but interspecies somatic cell nuclear transfer (iSCNT) faces challenges such as mitochondrial DNA incompatibility and potential issues with inbreeding.

Prospects of Germline Nuclear Transfer in Women With Diminished Ovarian Reserve

Antonia Christodoulaki et al.

Summary: Diminished ovarian reserve (DOR) is a challenge for assisted reproduction technology due to reduced quantity and quality of oocytes. Germline nuclear transfer offers a potential solution by allowing the generation of genetically related embryos. Different NT strategies could increase the number of available reconstituted embryos for DOR patients.

Cloning and Embryo Splitting in Mammalians: Brief History, Methods, and Achievements

Mohaddeseh Rahbaran et al.

Summary: Embryo splitting is a newly developed method in reproductive biotechnology, used in reproductive cloning studies and reproductive medicine research. By splitting embryos into individual blastomeres, genetically identical splitting can be achieved. This approach is widely employed in studying human diseases, treating sterility, embryo donation, and gene therapy.

Blastocyst-like structures generated from human pluripotent stem cells

Leqian Yu et al.

Summary: The study developed an effective three-dimensional culture strategy to generate blastocyst-like structures in vitro which resemble human blastocysts. These structures, termed 'human blastoids', are readily accessible, scalable, versatile, and perturbable alternative to blastocysts for studying early human development, understanding early pregnancy loss, and gaining insights into early developmental defects.
