iTRAQ-Based Proteomics Analysis of Rat Cerebral Cortex Exposed to Valproic Acid before Delivery

Jing Lin et al.

Summary: The study identified 79 differentially expressed proteins in the cerebral cortex of VPA rat model, mainly enriched at synapses and involved in energy metabolism, thyroid hormone synthesis pathway, and cytoskeleton. The dysregulation of these proteins may contribute to autism-like behavior in rats, as well as being associated with neuropsychiatric disorders. Increased expression of proteins related to energy metabolism suggests ASD may involve mitochondrial dysfunction that requires further investigation.

Proteomic Profiling of Cerebrum Mitochondria, Myelin Sheath, and Synaptosome Revealed Mitochondrial Damage and Synaptic Impairments in Association with 3 x Tg-AD Mice Model

Liming Shen et al.

Summary: Alzheimer's disease is the most common age-associated dementia with complex pathological hallmarks, and proteomics analysis revealed vulnerable subcellular components and differential protein expression between AD and WT mice; these findings suggest that early events of AD involve decreased energy metabolism, impaired neurotransmitter synthesis, and changes in cytoskeletal proteins.

Oxidative Stress in Autism Spectrum Disorder-Current Progress of Mechanisms and Biomarkers

Xukun Liu et al.

Summary: Autism spectrum disorder is a neurodevelopmental disorder with increasing diagnoses worldwide. Early diagnosis and intervention can improve outcomes. However, there are no clinical biomarkers for ASD. Recent studies have found a link between ASD and oxidative stress, and identified potential biomarkers to detect oxidative stress.

The complement cascade in Alzheimer's disease: a systematic review and meta-analysis

Saffire H. Krance et al.

Summary: The study found that concentrations of clusterin and complement component 3 were significantly higher in the cerebrospinal fluid of Alzheimer's disease patients, while C-reactive protein concentrations were elevated in peripheral blood. The results collectively support elevated complement pathway activity in AD, characterized by increased CSF clusterin concentrations and inconsistently reflected by peripheral blood proteins related to an AD diagnosis.

Plasma and Fecal Metabolite Profiles in Autism Spectrum Disorder

Brittany D. Needham et al.

Summary: The study used mass spectrometry methods to identify metabolic differences between children with ASD and typically developing children. The findings indicated differences in amino acid, lipid, and xenobiotic metabolism, implicating oxidative stress, mitochondrial dysfunction, hormone level elevations, lipid profile changes, and altered levels of phenolic microbial metabolites. These results suggest a link between metabolism, gastrointestinal physiology, and complex behavioral traits in ASD.

Oxidative stress marker aberrations in children with autism spectrum disorder: a systematic review and meta-analysis of 87 studies (N=9109)

Summary: The meta-analysis revealed that blood concentrations of certain oxidative stress markers were higher in children with ASD compared to healthy controls, while others were significantly reduced. The findings suggest a relationship between oxidative stress and ASD pathogenesis, indicating potential for early diagnosis using glutathione metabolism biomarkers.

Se-Methylselenocysteine (SMC) Improves Cognitive Deficits by Attenuating Synaptic and Metabolic Abnormalities in Alzheimer's Mice Model: A Proteomic Study

Xiubo Du et al.

Summary: The study found that Se-methylselenocysteine (SMC) can reverse the pathological phenomena of metabolic disorders, synaptic dysfunction, and oxidative stress in an Alzheimer's disease mouse model. After SMC treatment, the expression of related proteins was reversed, which may be the main mechanism of SMC intervention in AD.

The role of innate immune genes in Alzheimer's disease

Ana Griciuc et al.

Summary: Recent studies have highlighted the crucial role of the innate immune system and neuroinflammation in the pathogenesis of Alzheimer's disease, with a focus on microglial receptors CD33 and TREM2. Therapeutic approaches targeting CD33 and TREM2, such as gene therapy, small molecules, and immunotherapy, have shown promise in modulating neuroinflammation and potentially reducing neurodegeneration in Alzheimer's disease. Antibodies against CD33 and TREM2 have entered clinical trials and may provide a new avenue for treating Alzheimer's disease by targeting neuroinflammation.

Blood biomarker discovery for autism spectrum disorder: A proteomic analysis

Laura Hewitson et al.

Summary: Current research efforts are focused on identifying potential blood biomarkers for autism spectrum disorder (ASD) to determine risk or aid in diagnosis. Proteomic analysis revealed altered levels of certain proteins in the serum of ASD boys, suggesting the possibility of using a panel of proteins as a blood biomarker. Further investigation is needed to validate the protein biomarker panel for ASD in boys.

Fecal Microbiome Transplantation from Children with Autism Spectrum Disorder Modulates Tryptophan and Serotonergic Synapse Metabolism and Induces Altered Behaviors in Germ-Free Mice

Lu Xiao et al.

Summary: The study investigated the relationship between gut microbiota and autism spectrum disorder (ASD)-like behaviors by transplanting fecal microbiota from ASD and TD children into germ-free mice. The results showed that the microbiome from ASD children can lead to ASD-like behaviors in mice, different microbial community structures, and altered tryptophan and serotonin metabolism. Some microbiota may be related to tryptophan and serotonin metabolism.
