Single-strand DNA breaks cause replisome disassembly

Kyle B. Vrtis et al.

Summary: Research has shown that collisions between the replicative CMG helicase and nicks in DNA templates can lead to different forms of double-strand breaks, causing replisome disassembly.

Genome-wide mapping of human DNA replication by optical replication mapping supports a stochastic model of eukaryotic replication

Weitao Wang et al.

Summary: The study successfully mapped early-replication initiation events in human cells using the ORM method, revealing a consistent distribution of replication initiation with accessible chromatin enrichment. The results indicate that stochastic regulation of replication kinetics is a fundamental feature of eukaryotic replication.

Approaching Protein Barriers: Emerging Mechanisms of Replication Pausing in Eukaryotes

Maksym Shyian et al.

Summary: During nuclear DNA replication, the replisome needs to pass through and duplicate each chromosome's full length in each cell cycle. When encountering tight DNA-protein complexes, the replisomes slow down due to active fork pausing process with recognition of a protein barrier by an evolutionarily conserved Fork Pausing/Protection Complex (FPC). The FPC not only protects forks from collapse at protein barriers but also stimulates the DNA replication checkpoint and regulates topological transitions near the replication fork, contributing to genome integrity and various physiological functions in eukaryotic cells.
